Background and PurposeEsophageal Cancer (EC) is one of the six most common cancers in the world. Though a great development has been received in the diagnosis and treatment of EC in recent years, the malignant disease is still threatening people’s life and health. EC is typical of rapid deterioration, high metastasis, poor prognosis and high mortality. The aggression of the cancer cells into the surrounding tissue and the tumor metastasis happened in adjacent or distal tissues at the early period of cancer are the main reasons of poor prognosis. Tumor metastasis is a multi-step and complex process, involving multiple genes, and contains series of steps like invasion, survival in the circulation, growth in distal organs, and so on. The prognosis is closely related to the protein expression of tumor metastasis. Searching has provided a valuable target for the improvement of the prognosis, which is now becoming a popular topic in the research of cancer.Bone morphogenetic protein7(BMP-7)is a member of the TGF-βfamily, which exists in various body tissue.BMP-7plays an important role in the process of bone and cartilage development, promoting the growth of bones and cartilage and inducing ectopic bones, in addition, it participates in many kinds of biological functions, such as the occurrence of embryonic development and regulate cell proliferation, differentiation, apoptosis and so on. In recent years a large number of researches have reported that the expression of BMP-7are related with a variety of tumor occurrence, development and even metastasis, such as colorectal cancer, stomach cancer, myeloma, bone sarcoma, malignant melanoma, prostate cancer, breast cancer, renal cell carcinoma and so on. These reports of BMP-7have different effects in promoting and inhibiting the proliferation of tumor cells, but there has no explicit report about the effects of BMP-7on esophageal Squamous cell carcinoma at home and abroad.The study is to explore the expression of BMP-7in ESCC, with the purpose of finding out the relationship between the expression of BMP-7and cancer occurrence, deterioration, histological grade, clinical stage, invasion and the lymph node metastasis, So as to provide biological indicator for ESCC prognostic.Materials and Methods1Object of studyThis study enrolled56ESCC tissue of surgical resection samples at the second affiliated hospital of Zhengzhou University in2009.9~2011.7. All cases hadn’t had any preoperative radiotherapy, chemotherapy or other treatment. All the56specimens were statistically categorized according to the age, histological grade, tumor infiltration depth, lymph node metastasis and distant metastasis.2MethodsUsing SP detect the expression of BMP-7in56cases of ESCC and adjacent normal esophageal tissue, we analyzed the correlation of the BMP-7expression and ESCC with the clinical pathological characteristics.3Statistical MethodsThe data were analyzed by SPSS16.0software. The paired x2test and fourfold table chi-square test assay were applied for different groups comparison (α=0.05).Results1The expression of BMP-7in ESCCThe expression of BMP-7was mainly found in the cytoplasm and nucleus of cancer cells. The positive rate of BMP-7expression in ESCC was73.2%, higher than the normal controls by12.5%(P<0.001).2Relationship between ESCC and the expression of BMP-7The positive expression of BMP-7correlated with invasion, lymph node metastasis and distant metastasis (P<0.05), but not with gender, age and histological grade in ESCC(P>0.05)Conclusion1BMP-7is an excessive expression, which may played an important role in the process of Cancerous occurred and development in ESCC.2The positive expression of BMP-7correlated with invasion, lymph node metastasis and distant metastasis, which showed that BMP-7expression will enhance with the malignant degree of the cancer, and thus it indicates that the BMP-7expression was related with the prognosis of tumor.3BMP-7can be used as molecular markers for detection of esophageal squamous carcinoma. It is expected to be as a candidate molecular index for esophageal cancer prognosis evaluation.
