OBJECTIVE: Through the experimental study of cryopreservation of rabbit skeletalmuscle tissue pathology changes after limb replantation, To provide feasibility andpreliminary theory basis for replanting extremities after deep cryogenic treatment.METHODS: Take only30healthy New Zealand white rabbit, were divided intoexperimental group A and group B(A is slow-freezing and slow-thawinging,B isslow-freezing and slow-thawinging) and control group.The control group was madedirectly samples after amputated limb. Experimental group in dimethyl sulfoxideinjection, heparin saline water elution, after dealing with the slow freezing, the slowthawing of group A, group B fast thawing after replantation and reattach blood after6hours away from the amputated limbs, collect A specimen of skeletal muscle madewith the control group.RESULTS: General observation: after the experiment through blood group A muscle is ruddycolor, good texture, elastic, stimulating contraction. Experimental group B: through blood aftermuscle swelling significantly, texture is poor, weak elastic, stimulate no obvious shrinkage. HEstained pathological histology changes: the control group: normal muscle cell and the nucleusform, without the muscle fiber fracture, did not see obvious cavity formation; Experimental groupA: mild edema, muscle cells, part of the muscle fiber fracture, but the muscle cells than normalform, A small amount of inflammatory cell infiltration. Experimental group B: muscle cells,edema, muscle cells vacuolation in great quantities, a lot of muscle fiber fracture, there is a lot ofinflammatory cells infiltration. Control group: sarcomere neatly, Z line clear, mitochondrial cristae density, cell membrane integrity, no swelling, structure is normal. Experimental group A:muscle fibers can be clearly seen, and perfectly aligned with complete muscle membrane, musclefiber gap widened, mitochondrial cristae number decrease or disappear, mild swelling ofmitochondria, happened few mitochondria vacuoles, no fracture, mitochondria dissolved nomuscle fibers. Experimental group B: muscle fiber structure disorder, a lot of muscle fiber fractureand dissolved, muscle fiber gap widening, not normal mitochondria, mitochondrial clear vacuolesand fracture.CONCLUSIONS: The rabbit limb replanted6hours after slow-frozing in different ofthrawing, Skeletal muscle has a different degree of damage. According to thepathological slices of cell damage statistical analysis, control group and experimentalgroup had significantly statistical differences. Experimental group A and group B hadobvious statistical difference, the slow thawing smaller groups of skeletal muscleischemia-reperfusion injury, as cryoprotectants line from the broken body limbreattachment after deep cryogenic treatment provide A basis for the long-termintegrity of muscle tissue structure.
