BACKGROUNDThyroid carcinomas constitute the most common malignancies of the endocrine system Pathological types include thyroid papillary carcinoma (PTC), follicular carcinoma, medullary carcinoma and anaplastic carcinoma. Thyroid papillary carcinoma accounts for about60%-89%of cases worldwide and its incidence increases gradually. Iodine,estrogen and EEDS (Bisphenol A) may play an important role in the development of thyroid carcinoma. Therefore, we studied herein the effects of potassium iodine, estrogen and BPA, alone or in combinations, on the proliferation of thyroid papillary carcinoma cells (BHP10-3) in vitro. The present study might shed light on the role of these chemicals in the growth and provide the basis for the prevention and treatment of thyroid cancer.MATERIALS AND METHODS10-7mol/L potassium iodate;10-8mol/L BPA;10-8mol/LE2;10-7mol/L potassium iodate+10-8mol/L BPA;10-7mol/L potassium iodate+10-8mol/LE2;10-8mol/L BPA10-8mol/LE2Were added to the culture of BHP10-3cell,and ethanol(0.1%,V/V) was used as control group. Cell proliferation activity was examined by CCK-8colorimetric assays, and the cell cycles of the stimulated cells were analyzed by flow cytometer. The expression of cyclinDl protein was observed by western-blot, the expression of cyclinDl mRNA was observed by semi-quantitative RT-PCR.RESULTS1.The cell viability was significantly decreased in10-3mol/L, while the proliferation activity was obvious increased from10-4mo/L to10-10mol/L, and the maximum proliferation rates was80.2%in10-7mol/L(p<0.05).2.The proliferation activity was obvious increased from10-4mo/L to10-11mol/L, and the maximum proliferation rates was129%in10-8mol/L.(p<0.05).3.The cell viability was significantly decreased in10-3mol/Land10"4mol/L, while the proliferation activity was obvious increased from10-5mo/L to10-11mol/L, and the maximum proliferation rates was113%in10-8mol/L(p<0.05). 4.After stimulation with10-7mol/L potassium iodate;10-8mol/L BPA;10-8mol/LE2;10-7mol/L potassium iodate+10-8mol/L BPA;10-7mol/L potassium iodate+10-8mol/LE2;10-8mol/L BPA10-8+mol/LE2for481uhe proliferation rates of papillary thyroid carcinoma cells were higher than control group.and the combined effects were greater than they work alone(p<0.05).5.The proportion of Gl phase were less in the10-7mol/L potassium iodate,10-8mol/L BPA and10-8mol/LE2;10-7mol/L groups than the control group, while the proportion ofS phase were higher. Compared to the10-7mol/L potassium iodate,10-8mol/L BPA and10-8mol/LE2groups, the proportion of Gl phase were less in the10-7mol/L potassium iodate+10-8mol/L BPA.10-7mol/L potassium iodateI10-8inol/LE2and10-8mol/L+BPA;10-8mol/LE2groups, while the proportion of S phase were higher(p<0.05).6.Compared to the control group, after stimulation with10-7mol/L potassium iodate;10-8mol/L BPA;10-8mol/LE2;10-7mol/L potassium iodate10-7mol/L BPA;10-7mol/L potassium iodate+10-8moI/LE2;10-8mol/L BPA10-8mol/LE2for48h.cyclin D1protein and mRNA expression levels significantly increased in BHP10-3cells.CONCLUSION1.Potassium iodate can promote proliferation of the papillary thyroid carcinoma cells in a certain concentration range(K)-410-10mol/L),while the higher concentration(10-3mol/L) can promote cell apotosls.2. Estrogen his a positive ellect on the proliferation of BHP10-3eells.3.The proliferation of Bisphenol A on the papillary thyroid cancer was two sides. high conccntrations(10-3~10-4mol/L) is toxic, low concentration (10-5~10-10mol/L) can promote cell proliferation.4. Effects on promoting proliferation of thyroid papillary cells possibly by promoting the expression of CyclinDl cells, so as to accelerate the process of cells in Gl phase to S phase...
