Expression of TNFAIP8in human pancreatic cancer and effect of TNF-AIP8SiRNA on apoptosis and invasiveness in panc-1Objective:The incidence of pancreatic cancer was gradually rising in recent years, in which about85%occured in the pancreatic head. There are about0.227million people who are new pancreatic cancer patients. In China the death accident is the same of the occurrence rate. The pancreatic cancer is the most dangerous cancer. Tumor necrosis factor-a-inducible protein8(TNFAIP8or TIPE) family are very recently identified proteins and induced by TNF-a stimulation and by activation of transcription factor NF-κB. TNFAIP8family consists of TNFAIP8, TIPE1(TNFAIP8L1), TIPE2(TNFAIP8L2) and TIPE3(TNFAIP8L3), which share considerable sequence homology to regulate cell functions. TNFAIP8was originally indentified in human metastatic head and neck squamous cell carcinoma (HNSCC) cell lines.The expression of TNFAIP8is associated with enhanced cell survival and inhibition of activities of the apoptotic enzymes caspase-8and caspase-3. TNFAIP8aslo palys an important role in human cancers.Human non-small-cell lung cancer and breast cancer expressed the higher level than the health tissue. However, it is not known whether TNFAIP8takes a part in the progression of pancreatic cancer, and if so, what role it may play.Methods:Immunohistochemistry is used to test if TNF-AIP8is positive expression in human pancreatic cancer tissue. We use SiRNA to transfection the panc-1cells and Fluorescence microscope,Real-time PCR, Western blot analysis are used to examine the effects of transfection. MTT is used to test the multiplication capacity of the transfection cells and the panc-1cells. Transwell is used to test invasion ability of the transfection cells and the panc-1cells. Result:1. TNF-AIP8is positive expression in human pancreatic cancer tissue.2. The MTT find that the proliferation of the panc-1cell is much more higher than the SiRNA Treatment panc-1cells. The invasion of the panc-1cells is stronger than the SiRNA Treatment panc-1cells by transwell.Conclusion:TNF-AIP8expression plays an important role in the Pancreatic cancer. And if we find a good way to control TNFAIP8or to detect it, it will help us to find a new way for the treatment of the Pancreatic cancer. The second part:The Expression of TNF-AIP8in chronic pancreatitis and its clinical significanceObjective: To observe the effect of TNF-AIP8in the progress of chronic pancreatitis.Methods:Immunohistochemistry was performed to detect the TNFAIP8protein in chronic pancreatitis and health controls.Result: The results showed that a stronger staining of TNFAIP8protein was detected in chronic pancreatitis tissues compared with normal pancreas tissue.Conclusion:These results indicate that TNFAIP8may play a important roles in the progression of chronic pancreatitis.
