Objective:The problem purpose is that to observe the change of latency time and amplitude of SEP and MEP in SCs and OECs co-transplantation repairs spinal cord rat and the study of relativity about EP and BBB scores,for the sake of looking for the more impersonal,credible,quantitative appraisal methods of cell transplanation treats the effect of SCI rats,and appraises the methodological therapy.Method:1 Forty-eight SD rats were randomly divided into injury control group(A group),experiment control group(B group)and SCs and OECs co-transplantation group(C group),16 per set;2 With NYU spinal cord attacking implement,from 25mm drop,attacking T9 spinal cord and making the rat bring about middle degree injury.Cell transplantation group receive cell transplantation,ways is:with 'Hoechst' mark SCs and OECs suspension liquid,concentration is 50,000/ul,Transplanting cuts off end to spinal cord distance from when on contused wound area border 1mm spinal cord central line.The depth is that 1.75 mms,1.5 mms,1.0 mms,0.5 mms get along respectively,infuse SCs and OECs liquid about 1ul,every needle keeps a needle for 5 minutes.The experiment control group forming DF-12 culture solution pouring identical dosages in there being no serum in identical location;The injury control group is not on the march interfering with;3 The random chooses 8 rats in the group who allies self with transplanting from SCs and OECs,mark,inject a group into cyclosporin and contrast a group, 4 per set,cyclosporin gets a shot until the injecta starting hypo cyclosporin set from transplanting front one day,every time 10 mg/Kg,until drawing materials,contrast a group injecting identical dosages normal saline.4 Observation target:1)Evoked potentials:Before the recording spinal cord damage,the damage instantly,before the transplant and selects material front SEP and the MEP incubation period and the wave amplitude change situation.2)Hindlimb motor function BBB scores:Damages latter 2 day to the spinal cord, transplants the non-blood serum DF-12 nutrient fluid latter 2 day and SCs and OECs jointly transplants latter 2 day to carry on a time of BBB movement function grade,later each week will carry on an once,until drawing materials. Results:1 Immediately,after every animal of group is in SCI,the latent period of SEP and MEP appear prolong obviously,amplitude reduces obviously.Immediately after the wound,the latent period and amplitude of every group SEP and MEP is A group latent period(13.1±7.62)ms,amplitude(0.9±0.23)uV,B group latent period(11.8±1.70)ms,amplitude(0.9±0.12)Uv,C group latent period(6.6±1.20)ms,amplitude(0.94±0.11)Uv,respectively.2 After injury 6~12 weeks,discovering BBB motor function scores of SCs and OECs co-transplantation group was higher than experiment control group(P<0.01),After injury 12 weeks,BBB motion function scores of SCs and OECs co-transplant group do not see higher than injury control group(P>0.05). Whose value is injury control group(12.0±0.63)score,SCs and OECs co-transplantation group(12.2±0.81)score,respectively.3 After injury 15 weeks,SCs and OECs co-transplant group SEP and MEP do not see higher than injury control group(P>0.05),having recovering obviously comparing with experiment control group(P<0.01).Whose value is the latent period of injury control group SEP(5.0±0.42)ms,amplitude(1.8±0.18) uV;the latent period of experiment control group SEP(8.1±1.51)ms,amplitude (1.5±0.10)uV;the latent period of SCs and OECs co-transplantation group SEP(4.2±0.79)ms,amplitude(2.0±0.29)uV;the latent period of injury control group MEP(4.5±1.01)ms,amplitude(11.9±1.65)uV;the latent period of experiment control group MEP(6.3±1.72)ms,amplitude(10.3±1.27)uV; the latent period of SCs and OECs co-transplantation group MEP(3.8±0.50) ms,amplitude(12.5±1.47)uV;4 Wound last 3 weeks carry out SCs and OECs transplanting than wound last 6 weeks transplant recovery obviousness;After the wound 3 weeks SEP latent period is(2.2±0.19)ms,amplitude(2.3±0.19)uV;After the wound 3 weeks MEP latent period is(2.2±0.29)ms,amplitude(17.1±0.68)uV;The immune restraint being in progress cures,spinal cord injury rat in SCs and the OECs transplanting front and back to recover comparatively obvious.Before taking materials,treat with immunosuppressant SEP latent period is(3.0±0.71) ms,amplitude(2.1±0.62)uV,MEP latent period is(2.4±0.67)ms,amplitude (15.2±2.33)uV.Conclution:The result of evoked potentials and BBB scores shows,SCs and OECs co-transplantation have the certain effect to SCI rats the recovering of the hindlimb motor function and nerve conduction function.Early phase carries out SCs and OECs transplanting to the function of spinal cord rats recovering being more obvious,the immune restraint being in progress in the cell transplanting front and back is cured,beneficial to the rat motor functional recovering.EP change is able to reflect spinal cord conduction function damages degree,EP technology is able to be used for SCI determining the nature,fixing position diagnosing,the prognosis judgement.
