ObjectivesTo construct a rapamycin-eluting biodegradable ureteral stent with PLLA,PDLLA and rapamycin, and evaluate the feasibility and effectiveness of the stent inthe treatment of ureteral traumatic injury.Methods1. Smooth muscle cells were primary cultured by enzyme digestion, and thenwere exposed to rapamycin at different concentrations. The effectiveness ofrapamycin on the proliferation of smooth muscle cells was investigated by MTTdetection. We constructed a rapamycin-eluting biodegradable ureteral stent throughblending PLLA, PDLLA and rapamycin.2. The animal model was made throughinjuring mid-ureter by a hand-made explosion device, which had been provedeffective in our previous study. Both rapamycin-eluting biodegradable ureteral stentand double-J stent were surgically implanted into the ureters. By intravenouspyelography (IVP), radioactive renography and histological findings at different timepoints, we evaluated the feasibility and effectiveness of the rapamycin-elutingbiodegradable ureteral stent.ResultsWe successfully cultured smooth muscle cells in vitro, and the result of MTTconfirmed rapamycin could effectively inhibit the proliferation of smooth muscle cellsdepending on concentration. We also constructed the rapamycin-elutingbiodegradable stent, which contains2755ug of rapamycin. The results of IVP,radioactive renography and histological examination demonstrated that therapamycin-eluting biodegradable stent could be used to treat ureteral injurieseffectively and be applied to maintain the drainage with good biocompatibility. Nospecial stent-related complications were found during the experiment. Conclusions1. The rapamycin-eluting biodegradable ureteral stent could meet the demand ofclinical application with good mechanical and biological characters.2. Therapamycin-eluting biodegradable stent showed favorable therapeutic prospects in thetreatment of ureteral traumatic injury compared with double-J stent.
