The Expression Of Ep-cam And E-cad In The Gallbladder Carcinoma And The Relationship With The Invasion Ability

Objective:To investigate the expression of the epithelial cell adhesion molecule (Ep-cam) and the epithelial type calcium adhesion proteins (E-cad) in gallbladder carcinoma with invasion ability.Method:1. Gallbladder cancer cells in primary culture.2. Transwell experiments detect gallbladder carcinoma cell invasion ability.3. RT-PCR detect the expression of Ep-cam and E-cad gene in gallbladder carcinoma cells which have different invasion ability.4. Western-blot detect the expression of Ep-cam and E-cad protein in gallbladder carcinoma cells which have different invasion ability.Results:1. Primary culture method can breed passage of gallbladder carcinoma cells.2.Primary cultured gallbladder carcinoma cells have different invasion ability.(P<0.05)3. The expression of Ep-cam and E-cad are associated with the invasion ability of gallbladder carcinoma cells,the higher the Ep-cam expression,the higher the invasion ability(P<0.05); the higher the E-cad expression,the lower the invasion ability(P<0.05).4.The expression of Ep-cam and E-cad in gallbladder carcinoma cells, which have the Similar invasion ability, were not significantly different no significant difference.(P>0.05)Conclusion:1. There is a good correlation between the expression of Ep-cam\E-cad and the invasion ability of gallbladder carcinoma cells;The Ep-cam expression is associated with gallbladder carcinoma invasion was positively;The E-cad expression is associated with gallbladder carcinoma invasion was negatively. 2. The expression of Ep-cam and E-cad was negatively correlated in the gallbladder carcinoma cells.3.The combined detection of Ep-cam and E-cad expression in gallbladder carcinoma cells has some value to the evaluation of the invasion ability of gallbladder carcinoma...
