Objective:To investigate the expression of PD-L1 in gallbladder cancer and its effect on the invasion ability of gallbladder cancer cells.Methods: Collected paraffin wax specimens and clinical data of cancer tissues and paracancerous tissues of 37 cases of gallbladder cancer patients undergoing radical surgery for gallbladder cancer in Shaanxi Provincial Cancer Hospital and Xianyang Hospital of Yan'an University from January 2018 to December 2019.Using immunohistochemistry to detect PD-L1 expression in gallbladder cancer and analyze the relationship between its expression and clinicopathological characteristics;Silencing the expression of PD-L1 gene in human gallbladder cancer GBC-SD cells by small interfering RNA(siRNA).Then use quantitative real time PCR method to detect the expression of PD-L1 mRNA in GBC-SD cells;Western blot method was used to detect the expression of PD-L1 in GBC-SD cells;Transwell chamber method was used to detect the invasion ability of gallbladder cancer cells.Results: The positive rates of PD-L1 protein expressed in gallbladder cancer tissues and paracancerous tissues were 40.5% and 2.7%,The positive rate of PD-L1 protein expression in gallbladder cancer tissues is higher than that in paracancerous tissues(P<0.0001),The expression of PD-L1 in gallbladder cancer is related to tumor differentiation(P = 0.0392),vascular infiltration(P = 0.0029),and T stage(P = 0.0435);After PD-L1-siRNA was transfected into GBC-SD cells,The relative expression of PD-L1 mRNA was lower than that of NC group and si-NC group(P = 0.0005),PD-L1 protein expression was also relatively lower than that of NC group and si-NC group(P<0.0001),The number of cell migration in PD-L1-siRNA group was lower than that in NC group and si-NC group(P = 0.0018).Conclusion: PD-L1 has a higher positive expresssion in gallbladder cancer tissues,Silencing PD-L1 gene in gallbladder cancer GBC-SD cells can impair the invasion ability of gallbladder cancer cells.Therefore,PD-L1 may be used as a new target for gallbladder cancer treatment.
