| ObjectiveTo explore the expression of TLR4on colon cancer cell,futher investigating thebiological function of TLR4on human colon carcinoma cells.Methods1. The expression of TLR4was analyzed by RT-PCR and flow cytometry.2. The production of cytokine on HT29by the signaling of TLR4was tested byreal time-PCR3. Dendtrtic cell was co-culture with tumor supernatant.The secretion ofcytokine on Dendtritic cell by real time-PCR.Results1. TLR4is expressed on the surface of HT29cells. And LPS could not affect theexpression of TLR4.2.TLR4signaling promotes the production of tumor necrosis factor-αã€IL-23andCCL20.3.The signaling of TLR4on HT29could increase the expression ofimmunosuppressive factor of IL-10and IDO on dendritic cell.ConclusionsTLR4is expressed on human colon cancer cells.TLR4signaling could regulatethe secretion of cytokine on tumor, but also increase the expression ofimmunosuppressive factors. It promotes tumor immune escape and surveillance. |
