Objective To study the dynamics of the proportions of IL-17-producing cells andanalyze the main source of IL-17of mice infected with Schistosoma japonicum andthe mechanism of the biggest change of the IL-17-producing cells at different stagesafter infection. Method The leukomonocytes were prepared from livers of micewith S. japonicum infection at different stages (0w,3w,5w,8w,13w afterinfection). The proportions of IL-17-producing cells were determined by FACS. Liverand colon leukomonocytes were stimulated with SWA and SEA for48h in vitro, andthe proportions of the IL-17-producing cells were determined by FACS respectivelyResults The proportions of IL-17-producing cells increased significantly inleukomonocytes of livers from the mice infected with S. japonicum. TheIL-17-producing cells in livers included CD3~+CD4~+cells,CD3~+CD4-cells andCD3-CD4-cells. The IL-17-producing T cells (IL-17-producing CD3~+CD4~+cells andIL-17-producing CD3~+CD4-cells) accounted80%of the total IL-17-producing cellsand the proportion of the IL-17-producing CD3-CD4-cells increased significantlyinfection. The IL-17~+RORγt~+innate lymphoid cells which belong to theIL-17-producing CD3-CD4-cells began to continue to increase significantly since the5th week post infection. The SEA stimulation of leukomonocytes of livers and colonof mice in vitro significantly increased the proportion of the IL-17~+RORγt~+innatelymphoid cells (liver P<0.001; colon P<0.05) while the SWA stimulation ofleukomonocytes of livers of mice in vitro slightly increased the proportion of the IL-17~+RORγt~+innate lymphoid cells (P>0.05). Conclusion The IL-17-producingcells are significantly increased at different stages after S. japonicum infection,andthis ground cells include CD3~+CD4~+cells,CD3~+CD4-cells and CD3-CD4-cells. Asone ground of IL-17-producing CD3-CD4-cells,IL-17~+RORγt~+innate lymphoid cellsbegin to continue to increase significantly since the5th week post infection and thisphenomenon is caused by SEA of the pathological parts of mice infected withSchistosoma japonicum.
