Objective In order to research the purified combinational compounds of curcumin transformated by M.purureus went of curcumin and detect curcumin derivant from it> at the same time we study of Curcumin Metabolites on the expression of low density lipoprotein receptor in HepG2cell line.Methods①The strain M. purureus went was used to microbial transformated by solid fermentation. Detecting the thallus control and substrate control by means of TLC, HPLC to search curcumin derivant, Separated the extractive metabolites via Slica column chromatography,their compostion are analysed by LC/MS so that we can preliminary ensure the structure of curcumin derivant.②Utilize MTT to detect the effect of curcumin converted products on proliferation activity of HepG2cell and select appropriate concentration by result of MTT, we study its affections on the expression of mRNA and protein of Low Density Lipoprotein Receptor in HepG2Cell Line by Real-time PCRand western-blot.③Picking up LDL form blood plasma and signing with DiI to generate Dil-LDL, through detect the fluorescent light from intracellular to assess the effect of drugs on activity of LDLR.Result②here could be three kinds of curcumin derivants detecting by TLC and HPLC in total curcumin metabolites, the result of LC/MS show that their relative molecular mass is338,308and520,named CM1,CM2and CM3.②The result of LC/MS show that CM1is Desmethoxycurcumin, CM2is bisdemethoxy curcumin and CM3is the curcumin derivant whose phenolic hydroxyl is replaced by amino-ethylene glycol.③There is not significantly restrain to HepG2when the density of CM1,CM2and CM3reach15μg/mL,so choosing the concentration of5μg/mL,10μg/mL and15μg/mL curcumin metabolites to impact on HepG2,and they all can significantly promote the expression of Low Density Lipoprotein Receptor (P<0.05) in vary density degrees,and have certain concentration-response relationship, result of FCM show that three curcumin metabolites’ fluorescent light from intracellular are significantly increase activity of LDLR..Conclusion Monascus can transform the curcumin to three curcumin derivants in some conditions, CM1, CM2and CM3can regulate serum lipids and resistance atherosclerosis properties were proved to be enhanced, which can establish the foundation for us to screen new efficient antilipemic drugs.
