Objective:Gastric ulcer has a high incidence in clinical, with its recurrent features,which brings a great deal of pain to people’s body and mentality. The onlywestern medicine has more side effects, and withdrawals to recur. Chinesemedicine in the treatment of ulcer highlights its unique advantages, which hasbeen recognized gradually. This study is to observe the change of thepathology of the gastric mucosa and the effect of serum levels of tumornecrosis factor (TNF-α), superoxide dismutase (SOD) and catalase (CAT) onthe gastric ulcer of the rats with the Chinese medicine Huazhuo jieduyuyangjian, investigate the treatment of gastric ulcer mechanism of Huazhuojiedu yuyangjian, and provide experimental evidence for broader clinicalapplication of this prescription.Methods:After7days’ being adaptively fed,60weight220-250g male Wistar ratsare randomly divided into A, B, C, D, E, F six groups of10each, accordingto the random number table. Group A is the blank control group, and B groupis the model group, C group the western medicine group, D group traditionalChinese medicine high dose group, E group Chinese medicine dose group, Fgroup Chinese medicine low dose group. Raised in a normal environment, theblank control group of rats is fed with a normal diet. The remaining groups ofrats are fed in high wet and high temperature, with a normal diet, allowed todrink free20%honey water every other day, gavaged grease and white spirit.After14days, to make ulcer model, the groups are gavaged with anhydrousethanol in accordance1mL/100g, except the blank control group. The groupscontinued to be fed with high fat and sugar diet after the completion of themodeling. After7days each treatment group is given the drug intervention: the western medicine control group is given ranitidine suspension gavage; theChinese medicine treatment group is given traditional Chinese medicine theHuazhuo jiedu yuyangjian with different concentration gavage; the blankcontrol group and the model group are given with0.9%normal saline,2mldaily per rat. Drug intervention is ended after4weeks’ gavage. Each grouprats fast for24hours but water, then the rats are killed, and decapitate forblood, for the detection of serum TNF-α, SOD and CAT content. With aseptictechnique, open the abdominal cavity, and remove stomach of the rats, and cutalong the greater curvature, rush the stomach contents with0.9%saline, andflatten it, observe the ulcer situation and calculate the ulcer index, thenobserve the gastric mucosa histopathological morphology in the electronmicroscopy.Results:1The general condition of the rats observation:The blank control group rats have the white and Shiny fur, in good spirit,food intake and drinking more, with responsive and flexible action. The modelgroup rats are yellow fur, color dark, lack0f luster, unresponsive, poor spirit,malaise, lack of exercise, food intake and water intake reducing. The Chinesemedicine treatment group and the western medicine treatment of rats’ mentalstate improve significantly, increasing the amount of eating and drinking, thefur getting shiny, especially the high dose group of Huazhuo jiedu yuyangjianhave the most obvious improvement.2Gastric mucosa histopathological observation:In the Electron microscopy, the model group is in active, ulcer depth ofmuscularis more surface exudates; the western medicine group and theChinese medicine dose group are in the healing phase, the ulcers superficialcompared with the model group, visible freshmen gland and granulationtissue, and the ulcer is healing.3Detection of indicators:Comparison of Serum TNF-α levels: the traditional Chinese medicinegroups of different dose and the western medicine group are lower than the model group, and the difference is statistically significant (P<0.05); theChinese medicine high dose group are lower than the low-dose group andmoderate-dose group and the western medicine group, and the difference wasstatistically significant (P<0.05); the western medicine and traditional Chinesemedicine in the dose group are not statistical significant (P>0.05), but higherthan the traditional Chinese medicine low-dose group, statistically significant(P<0.05).Comparison of Serum SOD content: the model group are obviously lowerthan the control group, a statistical significant (P<0.05); the western medicinetreatment group are higher than the traditional Chinese medicine low-dosegroup, a statistical significant (P<0.05), lower than the traditional Chinesemedicine high-dose group, a statistical significant (P<0.05), close to thetraditional Chinese medicine moderate-dose group, not a statisticallysignificant (P>0.05); the traditional Chinese medicine high-dose group arehigher than the moderate-dose and low-dose group, a statistical significant(P<0.05); the traditional Chinese medicine in the moderate-dose group arehigher than the low-dose group, a statistical significant (P<0.05).Comparison of Serum CAT content: Compared with the model group, thetraditional Chinese medicine high-dose group and the western medicinetreatment group increase with a statistical significant (P<0.05); the traditionalChinese medicine high-dose group are close to the level of the westernmedicine group, not statistically significant (P>0.05); the traditional Chinesemedicine high-dose group are higher than the traditional Chinese medicinemoderate-dose and low-dose group, a statistical significant (P<0.05); thetraditional Chinese medicine moderate-dose and low-dose group are close tothe western medicine group, not statistically significant (P>0.05).Conclusion:Huazhuo jiedu yuyangjian can improve the general situation of thegastric ulcer model in rats, and its mechanism can be achieved by reducing themodel animal serum TNF-α levels and improving the content of SOD andCAT. By increasing the mucosal oxygen supply, improving the local microcirculation, promoting the regeneration of the gland and enhancing themucosal repair capacity, the healing of ulcers is promoted, while improvingthe quality of ulcer healing.
