Objective:By observing SA’s influence on hemisection of spinal cord injury rat hind limbs movementfunction, musculus gastrocnemius cross sectional area and diameter, this paper is to check theexpression change of Bax and Bcl-2in spinal cord tissue in order to lay a theoretical foundationfor molecular mechanism of treating spinal cord injury with SA.Methods:30SD adult rats are divided into three groups randomly. Normal Group (Group A) includes10rats without any treatment. NS Group (Group B) and SA Group also have10rats respectively.Stable right2-3hemisection of spinal cord injury is established in Group B and Group C with1.5ml NS or6mg/kg SA (dissolved with1.5ml NS) injected through vein30minutes before themolding. BBB grading and IP experiment are done before the surgery, first day, third day andfifth day after the surgery and first week, second week, third week and fourth week after. In thefourth week after the surgery,4%paraformaldehyde is used for perfusion fixation. Rightgastrocnemius and waist1-waist5segments of spinal cord are taken. Right gastrocnemiustissues, after paraffin embedding and HE stain after section, are tested with their cross sectionalarea using HMIAS-2000multi-functional color cell image analysis management softwares. Afterparaffin embedding and section, spinal cord specimens are dyed by Bax and Bcl-2immumohistochemical staining using BAC method. Immunohistochemical reaction positive cellnumber is calculated. The average optical density of Bax and Bcl-2immunohistochemicalreaction positive cells is tested using HMIAS-2000multi-functional color cell image analysismanagement softwares. SPSS13.0statistical analysis software is used for data processing. Datais shown in the(x|-±s) P<0.05is of statistical significance.Results:1. spinal function judgement: rats from Group B and C were deprived of their total hind limbmovement capabilities in very short periof of time (1day after the surgery). After3weeks, BBBgrading results began to have statistical meanings (P<0.05). IP experiment result in the first dayafter the surgery fell to about13°from82°. The angle began to rise with days going on. After4weeks, the results began to have statistical meanings (P<0.05).2. compared with Group A, right gastrocnemius cross sectional area and diameter of Group Band Group C decreased. The decrease of Group C was more obvious than Group B. Thedifference is more statistical meaning (P <0.05).3. immunohistochemical results showed that Bax and Bcl-2were expressed in each neuron. After 4weeks, positive neuron number and average optical density in both Group B and Group Cincreased compared with Group A. The difference is more statistical meaning (P <0.05). Baxincrease is more obvious in Group B than in Group C. Bcl-2increase in Group C is moreobvious than in Group B. The difference is more statistical meaning (P <0.05).Conclusions:1. SA is conducive for the movement recovery of hSCI rats.2. SA can help relieve shrank gastrocnemius atrophy of hSCI rats.3. SA’s protection of hSCI is relevant to Bax decrease and Bcl-2increase.
