Objective:To study the expression levels of XEDAR in the gastric carcinoma cell lines (SGC-7901,BGC-823and MKN-45)and gastric surface epithelium(GES-1);detect the relationship between miR-339and XEDAR and the gastric carcinoma cell lines in vetrio experimentMethods:Western-blot and SYBR green I real time PCR were performed to access the expression of XEDAR in different cell lines.The expression levels of XEDAR were verified by real time PCR experiment and Western-blot again after transfecting miR-339mimics.0.05levels was used for rejection of null hypothesis.Results:(1) Real time PCR results showed that the expression levels of XEDAR in gastric carcinoma cell lines was higher than gastric surface epithelium. Compared with GES-1, the expression level of XEDAR in SGC-7901,BGC-823, MKN-45was9.25,8.21and3.21respectively;(2)miR-339can be overexpressed by transfecting with miR-339mimics respectively.(3) Western blot results showed that the expression levels of XEDAR in gastric carcinoma cell lines was higher than gastric surface epithelium. Compared with GES-1. the expression level of XEDAR in gastric carcinoma cell was2respectively;(4)miR-339can be overexpressed by transfecting with miR-339mimics respectively. Compared to transfection negative control and the blank group Western blot results showed that the expression levels do not change after transfecting with miR-339mimics。Conclusion:(1) Real time PCR results and Western blot results showed that the expression levels of XEDAR in gastric carcinoma cell lines was higher than gastric surface epithelium.(2) After transfection with miR-339-5p mimics the expression levels of XEDAR has not changed. |
