Objective:The level of actin-binding protein (Fascin) and matrix metalloproteinases (MMP-2and MMP-9) expression in invasive pituitary adenoma were detected,and the may pathogenesis about invasive pituitary adenomas were analysed.Methods:Immunohistochemistry was used to check the expression level in72patients with pituitary adenomas (27cases of invasive and non-invasive45cases).Statistical analysis the differences expression in three protein between the two groups and the expression correlation among different clinical characteristics patien. In this study.it including33males and39females, the age distribution of18-74years old. with an average age of42.56years old. Invasive pituitary adenoma judgment in accordance with the Hard-Wilson pituitary adenoma grading. included27Invasive pituitary adenomas and45non-invasive pituitary adenoma. There exist25non-functioning adenomas and47endocrine function adenoma.All tumors in15cases up to3cm in diameter and over,the remaining57cases the tumor specimen diameter is less than3cm.All tumors were not receiving radiotherapy or chemotherapy before surgery and were confirmed pituitary adenomas after surgery by pathological examination.Analyze the results by use the Immune response integral (immunoreactive score, IRS) half quantitative method and SPSS17.0software.Results:1. Fascin protein expression Located in the cytoplasm, MMP-2and MMP-9expression were localized in the cytoplasm and nucleus. The positive expression were highly brown particles.The protein expression levels of Fascin protein, MMP-2and MMP-9in invasive group were higher than the non-invasive ones (t=3.03,t=333, t=2.02, P<0.05), it indicates there has statistically significant.2. There were no correlation between Fascin protein and MMP-2(rs=0.95,P>0.05), no correlation between Fascin protein and MMP-9(rs=0A.P>0.05), no correlation between MMP-2and MMP-9(rs=0.35.P>0.05)3. there were no correlation of Fascin protein, MMP-2and of MMP-9with gender and whether the tumor have endocrine function.but correlation with the tumor size (χ2=8.53, 11.31,12.46, P<0.05)Conclusions:The study suggest that the high expression level of Fascin protein, MMP-2and MMP-9may have a value to assess the invasiveness of pituitary adenomas. Fascin protein expression of MMP-2, Fascin in protein and MMP-9, MMP-2and MMP-9expression in invasive pituitary adenoma was not significantly different (P>0.05), three expression levels of different sex patients tumor with or without endocrine function there is no obvious correlation, but associated with tumor size, but because of the limited sample size of this experiment, the three invasive tumor cell growth process whether to play a synergistic role in its molecular mechanisms still need to be further clarified in the pituitary adenoma and invasion, although in most cases there are three high expression, but that might be possible with the participation of other factors, the specific mechanisms and factors between concerted mechanism to be further studied.
