Study On Toutongning’s Effects On The Expression Of C-fos And C-jun In The Rat Migrainous Model

Objective This study was performed to observe the effect of Toutongning on the expression of c-fos and c-jun in the migrainous model and evaluate the potential implications of Toutongning on migraine.Methods1. Sixty Wistar rats, in both genders, were divided into six groups at random:the normal saline(NS)group, the model group, the Flunarizine Hydrochloride group, the high dose of Toutongning group, the mid dose group, and the low dose group.2. According to the reference, the normal saline group and the model group were intragastric administration by physiological saline (0.02ml/g) every day for seven days. The Flunarizine Hydrochloride group were intragastric administration by flunarizine suspensions (1.25ml/g) every day for seven days. The Toutongning groups were intragastric administration by Toutongning suspensions respectively (5.2g/kg,2.6g/kg,1.3g/kg) every day for seven days.3. On the15th day,20min after administration, the model group, the Flunarizine Hydrochloride group and the Toutongning groups were injected subcutaneously nitroglycerin by lOmg/kg to make the migraine model.4. To observe the behavioral symptoms in each group.5. The expression of c-fos and c-jun was detected with the immunohistochemical method after copying the migraine animal model for3h.Results1.The behavioral symptoms in each group:except the normal saline group, other groups after modeling20min appeared reddish two ears scratching head frequently, and climbing the cage.The phenomena peaked in30-50min,and continued for1h. The Flunarizine Hydrochloride group,the high dose of Toutongning group and the mid dose group decreased gradually to the normal condition. The low dose group and the model group disappeared at2-2.5h.2. In leicaDM4000B, the positive cell was yellow. 3.The number of cells of c-fos and c-jun in the model group increased more significantly compared with that in the NS group(P<0.01).The expression of c-fos and c-jun in the high-dose Toutongning group and Flunarizine Hydrochloride group decreased compared with the model group(P＜0.01). The expression of c-fos and c-jun in the mid-dose Toutongning group decreased compared with the model group(P＜0.05). The expression of c-fos in the low-dose Toutongning group decreased, but there was no significant difference between the model group and the low-dose Toutongning group. The number of cells of c-jun in the low-dose Toutongning group decreased, compared with the model group(P＜0.05).Conclusion The results suggested that Toutongning could lower the expression of c-fos and c-jun which was induced by nitroglycerin and could inhibit the transmission of nociceptive pain information.
