Cancer has high mortality and threats human life, so researchers have long been committed to finding safe and effective anticancer drugs. Natural drugs have always been the important source of innovative drugs, and have unique advantages and potential in many disease areas such as cancer, inflammation, immunity. Medical plants are extensively distributed in China and we have much experience of application of herbal medicine, therefore it is imporntant to find natural antitumor drugs from traditional Chinese medicinal plants.Vitex is important medicinal plant and has been reported to exhibit a broad range of bioactivities, namely, antiinflammatory, anticancer. antibacterial,antioxidation. A large number of diterpenes, triterpenes and flavonoids, steroids have been isolated from Vitex species. The plant Vitex trifolia Linn. var. simplicifolia Cham is the genus Vitex of the family Verbenaceae. mainly distributed coastal areas of China. Its fruits (fructus viticis) are used as folk medicine in China for treatment of rheume, vertigo. Comepared to other species in Vitex, this plant was rarely investigated, only some scholars have studied its chemical composition and bioactivities.In this paper, we studied the chemical constituents in the methanol extract from the fruits of Vitex trifolia Linn. var. simplicifolia Cham collected in Wendeng, Weihai. Shandong Province. On the basis of spectroscopic methods, fourteen compounds were identified and they were elucidated as diterpenoids, flavonoids, triterpenes and sterols. Two labdan-type diterpenoids are new compounds. In addition, the results of bioactivities assay indicated that compounds vitrifolin A (1), vitexlactam A (2), vitrifolin B (3) had strong antitumour activities for A549cells, HepG2cells, Smmc-7721cells, and compounds5-hy droxy-3’,4’,6,7-tetramethoxyflavone (7), ayanin (9) had strong antitumour activities for A549cells. Meanwhile, We analyzed the best seperation conditions of compounds izalpinin (4), artemetin (5),5,3’-dihydroxy-7,4’-dimethoxyflavanone (6).5-hydroxy-3’,4’,6.7- tetramethoxyflavone (7),5,3’-dihydroxy-6,7,4’-trimethoxy-flavanone (8), ayanin (9) which were isolated from fructus viticis and optimized the extraction conditions of compounds artemetin (5),5-hydroxy-3’,4’,6,7-tetramethoxyflavone (7) by orthogonal experiment, their best extraction conditions were respectively five times amount of70%ethanol at60℃extracted three hours and the10-fold amount of70%ethanol at60℃extracted four hours.In summary, the thesis comprehensively investigated the chemical constituents and bioactivities of the fruits of Vitex trifolia Linn. var. simplicifolia Cham, which might be of significance for the exploitation of this specie and helpful reference for pharmacology and screening new medicine.
