Background: small cell lung cancer (SCLC) is the most frequently occurring cancers in theworld, and also the most common causes of cancer-related death in the world. Despite it’s towardmodest improvement, the outcome still extremely poor. Chemotherapy is the basis of treatmentin SCLC, and limited stage SCLC may have the chance of doing surgery, to extensive stageSCLC chemotherapy with or without radiotherapy is considered the standard treatment. So it isvery important to have further knowledge about the prognostic factors of SCLC. And the studyabout high risk patients and individual treatment is the hot point nowadays. Fli-1is transcriptionfactors and a member of the Ets gene family. Like other Ets proteins, Fli-1functions as both atranscriptional activator and repressor. As a proto-oncogene, Fli-1plays an important role inerythroleukemia development. And dysregulation of Fli-1is also observed in other malignancies,such as Ewing’s sarcoma, primitive neuroectodermal tumors (PNET). The detection of FLI-1intumor diagnosis has highly value in clinical diagnosis, so expression of Fli-1in malignant tumoris hot point in clinical studies.Objective: to investigate expression of Fli-1protein in small cell lung cancer, and analysisFli-1expression differences in small cell lung cancer and non-small cell lung cancer; explore therole of Fli-1in the diagnosis of small cell lung cancer and its clinical significance.Patients and Methods: Collected Small cell lung cancer patients underwent surgery at BethuneFirst Hospital of Jilin University and Beijing301Hospital from January2010to December2011,and used retrospectively analysis. Also we collected wax blocks and slices of lung tissue andadjacent normal lung tissue specimens. All patients’ postoperative pathological examinationconfirmed small cell lung cancer. A total of25cases of small cell lung cancer were included inthe study. Another Collected10cases of non-small cell lung tissue paraffin blocks. Fli-1antibody labeling of tissue samples, record Fli-1expression levels in different histological typesof lung cancer and expression intensity and expression in the staging of small cell lung cancerusing immunohistochemical methods of measurement results Wilcoxon test was used for statistical differences in the analysisResults:25patients with small cell lung cancer patients with a median age of58years old (44to78years old), and≥65-year-old patients were5(21%).15males and10females (male tofemale ratio of1.5:1); formulated in accordance with the American Legion (VALSG)installments:14cases are limited stage (54.2%), and11cases are extensive stage (45.8%). Fli-1antibody-positive cells were localized in the nucleus, Fli-1positive rate was20%in non-smallcell lung cancer, in small cell lung cancer and adjacent organizations Fli-1positive expressionrates were87.5%and0%(p=0.001); in limited stage and extensive stage Fli-1positiveexpression rate of84.6%and90.9%, respectively (p=0.006). Fli-1expression levels in small celllung cancer is significantly higher than in the adjacent tissues and non-small cell lung cancer,Fli-1expression significantly higher in the extensive stage than in the limited stage. InExpression intensity, Fli-1expression intensity in SCLC cancer tissue was significantly higherthan in the adjacent tissues (p=0.001); Fli-1in widely during expression intensity wassignificantly higher than the Bureau of the deadline, but the result has not statistically significant(P=0.083). Fli-1expression intensity in NSCLC was significantly lower than in SCLC (14and8),the results were statistically significant (p=0.001)。Fli-1expression levels in small cell lungcancer have no diffidence among different gender, different age stratified and the Ki-67expression levels.Conclusion: The study found that Fli-1expression levels in small cell lung cancer significantlyhigher, it can become a molecular marker for the diagnosis of small cell lung cancer;simultaneously Fli-1expression level is higher in extensive stage than in the limited stage, it canbecome a judge indicator of small cell lung cancer staging. And the gene in the progression ofthe SCLC played an important role, and may become a potential therapeutic target in SCLC, theresearch results will provide important clinical.
