Cancer is a major disease that seriously endangers human life and health. At present, the overall treatment of cancer patients has not been substantively improved. One of the key reasons is the unavailability of convenient method to detect cellular alterations in ultra-early stage of carcinogenesis processes, where genetic aberrations at nano-scale have not yet resulted in histological changes. The process of carcinoma formation involves step wise accumulation of genetic and epigenetic alterations in epithelia cells over a time period of many years. When the structural abnormalities corresponding atypical hyperplasia can be distinguished by optical micro-scope,it is already a relatively late event. From a cancer-research perspective, it is important to recognize the earlier stages of carcinogenesis that precede histological changes.Based on one-dimensional optical transmission within weak scattering and weakly disordered biological cellular system and its nano-scale sensitivity on fluctuations of refractive index, this project aims to develop an optical method and system for ultra-early detection of cancer. Mechanism of the mesoscopic light reflection spectroscopy for ultra-early detection of cancer is explored. Fetching and processing system on mesoscopic light reflection spectroscopy is established and has been applied to several samples for two-dimensional imaging based on disorder strength.According to mesoscopic light transport theory, the mechanism of mesoscopic spectroscopy for perceiving reflective index fluctuations at length scale of nanometers id revealed, and the feasibility of taking disorder strength as the optical fingerprint of a cancerous cell is analyzed. As the information catched by the fetching system of mesoscopic spectroscopy only reverse the one-dimensional channel comprehensive effect, seriously affected by surface relfection, depth-segmented partial-wave microscopic spectroscopy is put forward and localized mesoscopic light reflection spectroscopic system is developed. Simulation on depth-segmented detection and chacterization is done.Based on the above theory, Localized mesoscopic spectroscopic light reflection fetching and processing system was established. Through the data processing analysis of local mesoscopic reflection spectrum, we acquired the disorder strength image of reflection spectrum which reflected from the interest parts of the sample. This can effectively restrain the uninterested parts reflection spectrum’s influence, and is very favorable to the actual application.
