Objectives:1、To study the effects of HACC on the proliferation, invasion and metastasis of A549cells, and to explore their mechanism preliminaryly.2、To preparate the antineoplastic and hemostasis paster of HACC, and preliminary investigate its properties.Subject:1、Human non-small cell lung cancer A549cells;2、The antineoplastic and hemostasis paster of HACC.Methods:1、HACC was dissolved in1640culture medium, and set up the experimental groups of low,middle and high of the concentration of HACC. The HACC concentrations were set as follows:50μg/ml、100μg/ml、200μg/ml, the control group is1640culture medium. After seeded for24h, the Trypan blue method was used to analyze the cytoactive of A549; MTT assay was performed to study the inhibitory rate of HACC cells proliferation; and the expression of Matrix metalloproteinase9(MMP9) were detected by Western blot.2、HACC was dissolved in ultrapure water, and set up the experimental groups of different concentrations of HACC. The HACC concentrations were set as follows:1mg/ml、2mg/ml、4mg/ml. Put the different concentrations of HACC in the six orifice plate of cell culture, then place them into the-20℃refrigerator, when they became in blocks, removed them into the freezer dryer in order to mold. To observe the appearance of the paster, analysis the microstructure in the optical microscope, and set out density、water absorption rate and estimate the blood compatibility.Results:1. The Trypan blue dyeing showed that the viable rate of A549were more than95%, that fitted the requirement of experiment. The inhibitory effect of HACC on proliferation (P<0.05)in A549cells were treated by the HACC concentrations from50μg/ml to200μg/ml for24h were so obvious that comparing to control group, with the HACC concentration were increasing, the inhibition was gradually reinforced.2. The experimental groups of HACC concentration from50μg/ml to200μg/ml HACC could decrease the expression of MMP9in a dose dependent manner, comparing to the control group, it was significant (P<0.05).3.The suitable concentration of Comparing to the control group, with the concentration of preparating the antineoplastic and hemostasis paster of HACC was4mg/ml, the paster was white, the rate of pore-creating was high, the water-absorbing capacity was fine, and the blood compatibility was also all-right.Conclusion:1. The HACC concentration groups from50μg/ml to200μg/ml could inhibit proliferation, besides, they could inhibit the invasion and metastasis though restraining the expression of MMP9. It initially showed that HACC resisted tumor celles proliferating and metastasing through restraining the expression of Matrix metalloproteinases (MMPs).2. According the above-mentioned function, we used HACC to preparate the antineoplastic and hemostasis paster, the appropriate concentration of HACC is4mg/ml. The pore-creating rate of HACC antitumor hemostatic paster was high, the water-absorbing capacity was fine, and the blood compatibility was also all-right, the antineoplastic and hemostasis surgical dressing was worth researching and developing to put use in process of surgery.
