Objective To discuss the influence of phosphorylated STAT3on the model of acute pancreatitis associated with lung injury.Methods:The Kunming white, ICR, Balb/c of different strains of mice induced by intraperitoneal injection of caerulein to establish severe acute pancreatitis model,each strain has12.Caerulein(50ug/kg body weight)in0.9%NaCl at hourly intervals for12h、20h; The control animals received the same volume of0.9%NaCl without caerulein (0.2ml).The animals were killed after the last intraperitoneal injection。 The severity was evaluated in terms of amylase level, histological changes。To observate the differences between KM, ICR and Balb/c at12h,20h.To select the appropriate mice for the experiment. Findlly,blocking STAT3phosphorylation observed changes in tissue injury,18male mice were randomly divided into Group1:the control group (6), continuous intraperitoneal injection of0.2ml saline; group2:acute pancreatitis complicated with acute lung injury group (APALI6rats), to be intraperitoneal injection of cerulein (50ug/kg), dissolved in0.2ml saline; Group3rapamycin0.5mg/kg dissolved in DMSO in acute pancreatitis experimental group half an hour before injection (6).1h after the injection Pancreatic head, right lower lung for immunohistochemical examination, the body and tail of the pancreas and lung tissues for westemblots.Results:Balb/c12h、20h experimental group and KM20h experimental group with acute pancreatitis complicated with acute lung injury model succeed:pathology of pancreatic acinar appeared wide variability, and vacuolization acinar necrosis, a large number of infiltrating inflammatory cells。Interstitial and part of the alveolar space have more neutrophils, atelectasis. the gap widened alveolar。 KM12h and ICR20h experimental group were not successful. Between Balb/c12h and20h experimental group were not different (P>0.05).Between Balb/c12h and KM20h experimental group were not different (P>0.05).12h、20h experimental group and corresponding control group,serum amylase was significantly different in each strain.Conclusion Balb/C mice of drug response, and less drug use, saving time and costs for acute necrotizing pancreatitis model。Part Ⅱ To discuss the expression of phosphorylated STAT3on the model of acute pancreatitis association lung injury. Results Compared with normal control group, acute pancreatitis complicated with acute lung injury group pancreas, lung tissue phosphorylated STAT3strong positive expression, are found in alveolar epithelial cell, alveolar and inflammatory cells; phospho-STAT3protein expression increased; rapamycin treatment, pancreas, lung tissue expression of phosphorylated STAT3to alleviate. Only a small number of epithelial cells and inflammatory cells showed weak positive expression; westernBlot analysis of phospho-STAT3protein expression decreased significantly; pathological examination pancreas. lung injury significantly reduced,the extent of the three is basically the same.Conclusion phospho-STAT3is activated in acute pancreatitis complicated lung injury,by regulating the JAK2-STAT3activity, alleviate the injury of tissue.
