Effect Of Probiotic L. Casei Zhang On Preventing Rats From Development Of Type Ⅱ Diabetes And Its Improvement Of Immunity Response In Lype Ⅱ Diabetes Rats

Disturbed glucose metabolism and related chronic diseases have been a global difficulty, insulin resistance, type I and type II diabetes are of high incidence and considered as the second killer followed by cancer. The aim of this study is to investigate the preventive effect of L. casei Zhang on development of T2DM and ameliorative effect on immunological banlance of T2DM in rats. Especially, some immunity related cytokines and transcription factors were taken into account.In the present study, body weight, fasting and postprandial2h blood glucose levels, oral glucose tolerance, cytokines (TNF-α,IFN-γ,IL-10) and liver T-BET and GATA-3gene expression of rats were determined for evaluating preventive and therapeutic effect of L. casei Zhang on T2DM.Experimental SD rats were divided into four groups including control group, model group, preventive and therapeutic group. T2DM model was induced by2-week high fat diet plus with low dose of STZ injection (40mg/kg BW). Before injection, L casei Zhang preventive group rats were pretreated with a dose of4×109cfu/d-rat for3weeks. After injection, preventive and therapeutic group were treated with the same dose of L. casei Zhang for8weeks. At the12th week, all rats were sacrificed for index determination.Results showed that probiotic L. casei Zhang could prevent rats from T2DM development (P<0.01) but no therapy effects observed in fasting and postprandial2h blood glucose levels (P>0.05). Furthermore, preventive group exhibited significant lower TNF-a and IFN-y level and increased but not significant IL-10level (P<O.01, P<0.05and P>0.05) compared to T2DM model group after experimental model developed. In the end, therapeutic group exhibited significant decreased TNF-a and T-BET expression (P<0.05), increased IL-10expression (P<0.05) and no changes of IFN-y and GATA3expression (P>0.05) compared to T2DM model group. Changes of TNF-a, IFN-y and IL-10expression in preventive group were consistent with former data and liver T-bet and GATA3expression were respectively significantly decreased (P<0.05) and increased (P<0.05) compared to model group.All these results indicated that pretreated with L. casei Zhang for3weeks could prevent rats from development of T2DM.Although no significant therapeutic effect of L. casei Zhang on blood glucose was observed, L. casei Zhang played an important role in regulating Th1/Th2immunological banlance and inflammation allivation.Considering all these, probiotic L. casei Zhang exhibit potential application for development of T2DM related functional food.
