The Expression And Clinical Signifycanee Of MiR-451in Gastric And Colorectal Cancer

objectiveTo investigate the expression of miR-451in gastric cancer、 colorectalcancer and colorectal adenoma and its association with clinicopathologicfeatures in gastric and colorectal cancer.MethodsThe expression level of miR-451in18cases of gastric carcinoma andmatched non tumor adjacent tissue,25cases of colorectal cancer andmatched non tumor adjacent tissue and10cases of colorectal adenomatissue specimens was examined using Real time Polymerase Chain Reaction(RT-PCR).The relationship between the expression of miR-451and itsclinicopathologic features of gastric and colorectal cancer was analyzed.Adopt SPSS17.0covariance analysis software to carry on covariance to learna processing. Statistical analysis the gastric carcinoma and matched nontumor adjacent tissue was performed using the t-test to analysis of thevariance, Statistical analysis the colorectal cancer and matched non tumoradjacent tissue and colorectal adenoma tissue was performed using theone-way ANOVA to analysis of the variance, gastric and colorectal cancertissue and clinicopathologic features was performed using t-test to analysis ofthe variance. P＜0.05was considered statistically significant.ResultsThe expression level of miR-451was significantly lower in the tumortissues than that in the adjacent tissues(P＜0.05) which up-regulated in6cases，Down-regulated in12cases，Down-regulated rate66.67%（12/18）.Down-regulated miR-451expression was associated with the cell differentia-tion (P＜0.05) in gastric carcinoma patients. No significant association was found between the expression of miR-451and the status of age、gender、TNMstaging system、Borrmann type、depth of tumor invasion and tumor nodemetastasis stage；The expression level of miR-451was significantly lower inthe tumor tissues and adenoma tissues than that in the adjacent tissues（P＜0.01，P＜0.05）. which up-regulated in6cases，Down-regulated in19cases，Down-regulated rate76%（19/25）in colorectal cancer. up-regulated in3cases,Down-regulated in7cases，Down-regulated rate70%（7/10）in colorectaladenoma. Down-regulated miR-451expression was associated with the celldifferentiation (P＜0.05) in colorectal cancer patients. No significantassociation was found between the expression of miR-451and the status ofage、gender、Dukes staging system、depth of tumor invasion and tumor nodemetastasis stage.ConclusionsDown-regulated miR-451expression was may be involved in gastriccarcinogenesis and the development process.
