Objects:By using type1galanin receptor (GALR1) agonist M617to explore the effect of GALR1angonist M617on the glucose transportation in adipocytes of type2diabetic rats in the conditions of actvating GALR1, in order to understand the relationship among GALR1and GLUT4translocation, diabetes in adipocytes of exercise T2DM rats.Methods:Sprague-Dawley rats were fed with the high fat diet for eight weeks. The rats over200g were treated by one injection of streptozotocin (40mg/kg, i.p.) to establish type2diabetic models. The model rats were randomly divided into four groups. Rats from diabetic sedentary control group (n=10) and diabetic training group (n=10) were injected by saline vehicle (0.3mL/d, i.p.). Whileas rats from both diabetic sedentary drug group (n=10) and diabetic training drug group by M871(3μmol/kg/d). Twenty healthy rats were randomly selected into sedentary (n=10) and exercise control (n=10) groups and were injected by saline vehicle (0.3mL/D, i.p.) too. Rats from the three exercise groups swam (lh/d) after the injection for20days consecutively. Body weight of all rats was measured by electronic scale every week and blood sugar level was determined using a Blood Glucose meter. The insulin sensitivity of the tested rats was investigated by the euglycemic clamp tests, fasting serum insulin, galanin levels and GLUT4content in adipose homogenate by ELASA kits, GLUT4contents in adipocyte membranes by Western blot method, GLUT4mRNA levels of adipose tissue by Real-time PCR.Result:The results showed that blood glucose and insulin levels decreased significantly (p<0.05), but galanin content and quantity of GLUT4translocation in plasma membrane of adipocytes increased significantly (p<0.05) in both drug groups compared with each diabetes control. Besides, blood glucose and insulin levels decreased significantly (p<0.05), whileas, galanin content and quantity of GLUT4transportation in plasma membrane of adipocytes increased significantly (p<0.05) in both diabetes exercise groups compared with each diabetes sedentary group too.Conclusions:GALR1specific agonist M617promoted insulin sensitivity and increased GLUT4translocation in plasma membrane of adipocytes, which improved the ability to utilize glucose in adipocytes. These results suggested that galanin may regulate glucose metabolism by promoting GLUT4translocation from intracellular membranes to plasma membranes through GALR1in adipocytes.Exercise reduced blood glucose level and improve insulin sensitivity by accelerating the translocation of GLUT4or improve the activity of GLUT4. The experiment shows that the GALR1specific agonist maybe a new kind of drug to be used to deal with T2DM.
