Rhizoma Sparganii is a well-known traditional Chinese herbal medicine which originates from the rhizome of Sparganium stoloniferum Buch.-Ham.. It is widely distributed in Northeast, North, East, Southwest and other regions of China. It is reported that that Rhizoma Sparganii contains the volatile oils, flavones, organic acids, phenylpropanoids and steroides. Pharmacological studies have certified that it prossessed protective effects on cardiovascular and cerebrovascular systems. It is also applied clinically as hepatoprotective and anticarcinogenic agents. Rhizoma Sparganii is used to treat qi and blood stagnation, amenorrhea, abdominal pain and congestion after postpartum, food stagnation, bruises and other diseases. So far, the major active components of Rhizoma Sparganii have not been clarified.Based on the results of the in vitro active screening, the lowest polarity fraction of total extract of Rhizoma Sparganii showed significant effect. It could significantly inhibit the cell growth of Bel-7402at10μg/mL, and inhibited the cell growth of ECV304at lμg/mL. It could also observe an obvious effect on PMNs and ECV304at10μg/mL, and on thrombocyte and ECV304at1μg/mL and0.1μg/mL, respectively. In order to identify the active components, an activity-guided isolation was carried out, and1H-NMR, and13C-NMR spectroscopic methods were used to elucidate the structures. Furthermore, HPLC and MALDI-MS/MS methods were used to comprehensively identify the structure types of the active fraction, which was finally elucidated to be the phenylpropanoid types of compounds.According to the results of in vitro screening, it proved that Rhizoma Sparganii has property of anticancer, and has effects on cardiovascular and cerebrovascular systems, just like what has reported before.This paper also established the fingerprint of Rhizoma Sparganii using high-performance liquid chromatography (HPLC) method. The HPLC fingerprint showed13characteristic common peaks, among which8peaks were recognized. Twenty-one batches of Rhizoma Sparganii were evaluated by the established method. And the similarities of the whole samples were over0.89. Organic acids and phenylpropanoids were the major compounds of Rhizoma Sparganii. The method is repeatable and reliable, and it is capable of effectively controlling the quality of Rhizoma Sparganii.
