Objective To investigate the effect of PDGF-α receptor silence on proliferation and apoptosis of human lens epithelial cell in vitro, so that experimental data was provided for the treatment of posterior capsule opacification.Methods Lens epithelial cells were cultured in vitro and PDGFR-α receptor antisense oligonucleotides was transfected into cells by cationic liposomes,the morphological changes of SRA01/04was observed under inverted microscope. PDGF-α receptor antisense oligonucleotides has effect on SRA01/04cells24h~72h,and cell proliferation was detected by MTT experiment. PDGF-α receptor antisense oligonucleotides has effect on SRA01/04cells48h,and cell cycle and apoptosis was detected by Flow Cytometer, cell apoptosis by Hoechst33258, and PDGF-α receptor expression changes of PDGFR-αASODN role after48h was detected by Reverse transcriptase Polymerase Chain Response(RT-PCR).Results1. Most cells of the experimental group become shrinkage, roud, and even fragmentation, the dead cells float in the culture medium, and part of live cell division phase transition, attachment is not strong, the fuzzy boundaries of the cell body even lost their original shape after treated with PDGFR-αASODN48h.Compared with the low concentration of the drug group, the high concentra- tion of the drug group stronger effect on inhabita-tion.2. PDGF-α receptor antis-ense oligonucleotides has effect on SRA01/04cells24h~72h, and the inhibition of SRA01/04increased with time,compared with the control group and misse-nse oligonucleotide group,the experimental group stronger effect on inhabitation, and there was significant difference (P<0.05).3. PDGF-a receptor antisense oligonucleotides could induce apoptosis SRA01/04cells,After treated with PDGFR-aASODN48h,;the SRA01/04cells apoptosis ratio of experimental group were (3.22±0.25)%and (5.29±0.27)%,showing significance with control group (0.75±0.67)%and missense oligonucleotide group (1.46±0.60)%(P<0.05) percentage of cells in G1phase in experimental group were (53.31±1.30)%and (59.98±0.95)%, showing significance with control group (47.73±1.18)%and missense oligonucleotide group (49.48±1.09)%(P<0.05).4After treated with PDGFR-aASODN48h,Hochest33258staining Showed that:the experimental group nuclei pyknotic hyperchromatic nuclei, dense apoptotic cells compared with control group increased.5.After treated with PDGFR-aASODN48h, the experimental group of PDGF-areceptor has weak expression in SRA01/04.Conclusions1、PDGF-α receptor silence could inhibit human lens epithelial cell proliferation and induce apoptosis.2、PDGF-α receptor antisense oligonucleotide silence the PDGF-a receptor expression in the SRA01/04.3、PDGF-a receptor play an important role on proliferation in human lens epithelial cells.
