| Objective:To study the effects of metformin on the leptin〠resistinã€adiponectin mRNA and protein expression in serum and adipose tissue of obstructive sleep apnea syndrome (OSAS) Chinese fragrant swine model.Methods:Twenty-three Chinese fragrant swines were randomly divided into control group(six)ã€model group(six)ã€metformin intervention group(six). obstructive sleep apnea hypopnea syndrome (OSAHS) animal model were established through injecting the contents of medical ice pack into the pharyngeal palate of Chinese fragrant swine in the model group and metformin intervention group.the metformin treatment group were fed with metformin0.2g·kg-1·d-1, the control group and OSAS model group were_fed with equal volume of normal saline Once a day. Each group were sacrificed after12weeks and drawn materials. Each group were detected Resistin, Leptin level in serum using ELISA method, Using polysomnography respiratory monitor to monitor, leptin, resistin,adiponectin gene and protein expression were assayed respectively by RT-PCR and Western-blot method in omental adipose tissue.. Results:Compared with the control group, Leptinã€resistin mRNA and protein expression gray value in serum and adipose tissue were rised, on the contrary, adiponectin mRNA and protein expression gray value in serum and adipose tissue were declined,the difference was statistically significant (P<0.01). Compared with the OSAS model group, after metformin treatment, sleep apnea hypopnea index (AHI) decreased, the lowest blood oxygen desaturation (SaO2) increased (P<0.05) Leptinã€resistin mRNA and protein expression gray value in serum and adipose tissue were dropped (P<0.05), adiponectin mRNA and protein expression gray value in serum and adipose tissue were rised (P<0.05).Conclusions:Metformin can alleviate insulin resistance;can make leptinã€resistin mRNA and protein expression restrained and adiponectin mRNA and protein expression increased, to play its role of intervention in OSAS. |
