Objective To study the antineoplastic activity, induce-cell apoptosis activity and its mechanisms of Chinese medicinal FFJZ. To produce the scientific evidence for developing and manufacturing new antitumor drugs.Methods1. The suppressive effect of FFJZ on the proliferation of Raji tumor cells were investigated in vitro by using MTT colorimetric method.2. The apoptosis of Raji induced by FFJZ was studied by Hoechst-PI and Annexin Ⅴ FITC/PI, DNA agarose gel electrophoresistechnique.3. There were19cases in treatment group,treated with FFJZ plus chemotherapy, There were21cases in control group, treated with chemotherapy. The clinicial therapeutic effectiveness was observed between two groups.Results1. FFJZ inhibited markedly the proliferation of Raji cells, with dose-dependent relationship.2. After treatment with FFJZ, the Raji cells presented some typical morphologic features and super-microstructural changes of apoptosis, including cell shrinkage, cytoplasm concentration, nclius forming outwardly acute angle promineney,chromatin concentrating on karyotheca or forming meniscus, nuclear condensation, nuclear fragmentation and formation of apoptotic bodies. The apoptotic rate of Raji was increased after FFJZ treating for72h. DNA agarose gelelectrophoresis showed characteristic "DNA ladder" pattern.3. The effective rates of treatment group and control group are71.4%and75.8%, showing statistical no difference (P>0.05), Days of keeping CR shows statistical difference (P<0.05), There are statistical difference between two group of side effect (P<0.05).Conclusion1. FFJZ has very strong antitumor effect in vitro and in vivo. FFJZ can inhibit the proliferation ofRaji cells invitro.2. FFJZ can not increased the objective response rate of lymphoma, but can conspicuously synergy the chemotherapy and decreased adverse events.
