Objective:1. Establish experimental models of acute myocardial ischemia/reperfusion in rats by ligating coronary artery with capsule.2. Observe the protective effect of ischemic postconditioning(IPost) to acute myocardial ischemia/reperfusion in rats with the method of repeatedly drawing-aerating the capsule3. Explore the possible mechanism.Methods:In this experiment in vivo myocardial ischemia and reperfusion model in rat was established by ligating coronary artery with capsule, and IPost was produced through repeatly drawing-aerating the capsule. SD rats were randomly divided into3groups: Sham operation group, ischemia/reperfusion group (I/R group), ischemic postconditioning group(Ipost group). Animals were sacrificed at the180mins after reperfusion to determine cardiac functional parameters, plasma myocardial enzymes, cell apoptosis index and the myocardial infarct size. Western blot was used to analyze the expression of Bcl-2、Bax、Caspase-3in myocardium.Results:The result show that the methods establising MI/RI model in rats by ligiating coronary artery with capsule and producing Ipost by repeatly drawing-aerating the capsule were convenient and speedy. Compared with I/R group, the heart function parameters significantly increased(P<0.05), while the levels of plasma myocardial enzymes, apoptosis index and myocardial infarct size significantly decreased(P<0.05). The expression of Bcl-2in myocardium in the IPost group were significantly higher than that in the I/R group, while Bax and Capase-3were all significantly lower(P<0.05). Conclusion:1. It is fast and convenient to imitate IPost with the method of ligating coronary artery with capsule.2. IPost can alleviate myocardial ischemia/reperfusion injury, reducing myocardial apoptosis and infarct size.3. The protective mechanism is possible associated with the activation of RISK signaling pathway which inhibits the open of mPTP.
