Rhizoma Polygonati Odorati (Fragrant Solomonseal Rhizome; Yu Zhu) is the dried rhizome of Polygonatum odoratum (Mill.) Druce. It is a common Chinese medicinal herb which has a long history of indigenous use and well-known traditional Chinese medicine used for removing dryness, promoting secretion of fluid and quenching thirst in China. In order to develop the Polygonatum odoratum, a microwave-assisted procedure for the extraction of Yuzhu polysaccharides was established and prepared Polygonatum doratum polysaccharide-iron (III) complex (POPFe).Part I Optimization of polysaccharides extraction from Polygonatum odoratum using response surface methodologyObjective:In this study, a microwave-assisted procedure for the extraction of Yuzhu polysaccharides was established. And the antitumor activity of the polysaccharides was analyzed.Methods and results:1. A Box-Behnken Design (BBD)(4factors and3levels) was employed to optimize the microwave-assisted extraction (MAE) conditions. The results indicated the optimum extraction conditions were as follows:temperature of57℃, microwave power of300W, extraction time of10min and water/solid ratio of23:1. Using a short application of microwave, the mean experimental yield of polysaccharides was17.49%, which was well in close agreement with value predicted by the model.2. We studied the anti-tumor in vitro experiment of crude polysaccharides on Bcap-37, Pan-1and A549by using MTT method. The conclusion was:all of them have effect on Bcap-37, Pan-1and A549, but the inhibition ratio are variously.3. The polysaccharide was purified by activated carbon and enzyme-TCA, and then separated by the DEAE-52and SephadexG-100.Conclusion:The microwave-assisted procedure was easy and reasonable, and the polysaccharides had the anti-tumor activity on Bcap-37, Pan-1and A549. PartⅡ Study on preparation and chemical characterization of IRON (Ⅲ)-Polygonatum doratum Polysaccharides complexObjective:To prepare Polygonatum doratum polysaccharide-iron (Ⅲ) complex (POPFe) and study some physicochemical properties of POPFe.Methods and results:1. POPFe was synthesized by the neutralization of a FeCl3carbohydrate solution. Orthogonal design was used to optimize the preparation process. The content of iron (Ⅲ) in POPFe was determined with o-phenanthroline spectrophotometry and iodometry. The optimization preparation conditions were found to be a ratio of four polysaccharides aqueous solution to the sodium citrate solution, temperature70℃and pH9. POPFe showed the qualitatively identified reactions of Fe (Ⅲ). The content of Fe (Ⅲ) in POPFe is30.13%.2. Surface structure of POPFe was studied by Infrared spectroscopic technique. The deoxidization of POPFe was tested with colorimetric analysis. The complex was stable at pH3~12without precipitation and dissociation. At37℃, Fe (Ⅲ) in the complex was completely reduced to Fe (Ⅱ) by ascorbic acid in about5hours.Conclusion:POPFe could become higher bioavailability and nutrition iron-supplementary.
