Objective:The influence Of the electroacupuncture on the aged rats’spatial memory-was studied through the observation on aged AD rats’behavior awaking system and the test on neurotransmitter content of DAn5-HT\NE.etc of3groups(the health rats group, the electroacupuncture group and model group) hoping to provide a new method to treat AD.Methods:Morris water maze was used to screen natural aging rats comparing with young rats of3-6months old. The aged rats were divided into2groups, the upper limit is the mean of young rats group’s average escape delitescence which added2times standard deviation,the lower limit is the mean which added1time standard deviation. The mean which was greater than the upper limit was AD group, which was less than the lower limit was health old rats group. The old rats whose incubation period between the upper and lower limit was eliminated.The AD rats group was divided into electroacupuncture group and model group with16rats in each group. Rats in the electroacupuncture group was stimulated on DU20and KI1after being fastened while rats in the other two groups were only fastened without any treatment. After treatmentmorris water maze was used to train and test the spatial memory ability of old rats in each group. Spatial memory includeded spatial reference memory and spatial working memory.The information swimming time was the research indicator of spatial reference memory. The choosing swimming time was the research indicator of spatial working memory.Then the camera would continuously record the awaking time and upright numbers of old rats in12hours(19:00-07:00).meanwhile testing monoamine neurotransmitter content of DAn5-HT, NE and5-H1AA.etc.Results:1. Spatial memoryComparing with the model group.both the information swimming time (22.94±4.36) and the choosing swimming time (26.90±3.39) of the-electroacupuncture group were obviously reduced with a significant difference from the model group’s information swimming time (27.66±3.50)(P<0.01) and the choosing swimming time (30.54±3.35)(P<0.05)2.Awaking abilityTo continuously oberve the awaking state of the rats at night12hours (19:00-07:00) after all the treatments the activity of electroacupuncture group was significantly increased at night comparing with the model group.The awaking time (453.3±28.1) of electroacupuncture group was obviously extended comparing with the model group (380.0±28.2) as well it also had a significant difference from the model group (P<0.001).Upright times of electroacupuncture group (6.90±3.39) was obviously increased with a significant difference from the model group (15.1±2.7)(P<0.001).3. Frontal lobe neural transmitters content.The frontal lobe neural transmitter content of DA、5-HT、NE、5-H1AA were tested after all other indicators have been tested.The electroacupuncture group’s neural transmitter content of DA(2.76±1.99),5-HT(0.57±0.22),NE(0.51±0.27).5-HIAA(7.30±1.51)were higher than the model group of DA(1.20±0.89),5-HT(0.38±0.11), NE(0.32±0.12).5-HIAA(4.19±1.14) and all the content had a significant difference from the model group(P<0.01or P<0.05).Conclusion:To stimulate with electroacupuncture on DU20and KI1could signi-ficantly increase old AD rats’neurotransmitter content of DA、5-HT、NE and im-prove the awaking state as well as work memory ability of old AD rats. The mech-anism is to promote the function of neurons increase the release of monoamine neur-otransmitters activate low function of neurons. Electroacupuncture started and main-tained the sleeping-awaking system of AD rats so that to improve the function of the learning and working memory.
