Objective:To explore the effect of mice serum containing kidney-nourishing herbs on the IVM of human immature oocyte.Method:The110obsolete immature oocyte complexes (COCs) supplied by Assisted Reproduction Center in the Second Clinical Hospital affiliated to Chengdu University of Traditional Chinese Medicine were randomly grouped of5group, i.e. the1,3,5groups with mice serum containing kidney-nourishing herbs, the blank mice serum group and the hormone (FSH-LH) group, which were placed to the HTF culture solution added to the No.1, No.2, and No.310%mice serum containing kidney-nourishing herbs,10%blank mice serum and10%blank mice serum containing FSH-LH with75IU/L respectively. After cultured24-72h with microscope method, using the inverted microscope, we observed and calculated the first polarbody (PB1)(i.e. maturating rate) of oocyte in vitro. Then, we performed intracytoplasmic sperm injection (ICSI) to the mature oocyte in culture solution and observed the fertility rate and cleavage rate after24h. With euzymelinked immunosorbent assay (ELISA), we determined the concentration of HLA-G in culture solution.Results1. There was no significant difference in the maturing rate, fertility rate and cleavage rate during the IVM of immature oocyte in the No.1mice serum containing kidney-nourishing herbs and the No.5mice serum containing kidney-nourishing herbs compared with the blank mice serum group.2. Compared with the blank mice serum group, the No.3mice serum containing kidney-nourishing herbs had significant difference in the maturing rate and fertility rate during the IVM of immature oocyte3. There was no significant difference in the cleavage rate during the IVM of immature oocyte in the No.3mice serum containing kidney-nourishing compared with the blank mice serum.4. There was no significant difference from the GV to the GVBD stage of the human immature oocyte in the No.1, No.2, and No.3mice serum compared with the blank mice serum group.5. With regard to the determination of the concentration of sHLA-G, the value was lower in No.5mice serum containing kidney-nourishing herbs (compared with the standard value); though the value of the No.3mice serum was higher, but there was no significant difference compared with the blank mice serum group.ConclusionIn addition to stimulating the growth and development of human immature oocyte the NO.3mice serum containing kidney-nourishing herbs mainly promoted the maturation of oocyte nucleus. The expression and its level of the concentration of sHLA-G in embryonic fluid at cleavage stage might correlated with kidney-nourishing herbs, however, the effect of the kidney-nourishing herbs on improving the quality of oocyte was not obvious.
