Objective ①Compare the maturation rate and maturity time among the different group.Analyze the effect of HCG matueation in vitro.②Appropriate time of mature oocytes will be calculated in each experimental group in order to create the conditions for the followup training. ③The fertilization rate, cleavage rate and high quality embryo rate will be compared among each group to investigate the effect of HCG reflect on eggs quality and the potential of embryo development. Methods ①All the eggs were divided into four groups according to the concentration of HCG. We compared different concentration of HCG on oocyte maturation rate and mature time and calculated the optimal incubation time of each group.②The fertilization rates, cleavage rates, high-quality embryo rates were compared among the groups.Compare the fertilization rate, cleavage rate, high-quality embryo rate among the groups.Analyzed the different concentration of HCG and incubation time on the effect of eggs quality and compared the difference between the in vitro maturation and in vivo maturation. Results ①The maturation time of the oocytes in the four experimental groups was 24 hours, 18 hours, 18 hours, 12 hours; The corresponding maturation rate was 81.82%,82.24%, 81.55%, 81.19%. There was no statistically significant difference in the four groups. ② The high-quality embrys rates were 28.44% and 29.25% and significantly increased than other two groups when the HCG concentrations were 75IU/L and 150IU/L.The group which oocytes were maturated in vivo has the best high-quality embryos rate(48.24%). ③ The high-quality embryo rate of HMG group was better than HCG group(38.18% VS27.83%).Conclusion ①Adding HCG or not does not affect the oocyte maturation rate in vitro.②Medium supplemented with HCG will cut down the time of maturation in vitro.③The results are affected by the concentration of hormone and the culture time in vitro.④The quality of eggs matured in vivo are better than which maturated in vitro.Figure21,Table9, Reference 105...
