Objective:Research on the phase characteristic and the mechanism of electric-acupuncture on the delayed circadian rhythm.Method:Syrian golden hamsters were synchronized to normal light/dark cycle, then delayed circadian rhythm models were made by11days of serial8-hour delays of light/dark cycle every2days. Contrasted the effects of electric-acupuncturing both " Bai hui (DU20)" point and "Changqiang (DU1)" point after delayed light/dark cycle at different time.Aslo.the level of PER1protein, CKIε and phosphorylation of PER1protein in SCN were observed through western blotting at ZT12.Results:Firstly, the spontaneous activites of syria golden hamsters shifted backward after delayed light/dark cycle.Their average shift extent of different groups at different time was from116.75to378.75minutes. Secondly, acupunctured at ZT0.ZT4and ZT16.the time of regulating circadian rhythm was less than bundling group.however.no defference in statistics. Acupunctured at ZT20. the time of regulating circadian rhythm was close to bundling group. The outstanding effective of regulating circadian rhythm was observed in electric-acupuncture group at ZT8and ZT12. Only when acupunctured at ZT12. outstanding effective of increasing activities was observed. Thirdly, at ZT12electric-acupuncture significantly promoted the expression of CKIε and phosphorylation of PER1in the SCN. while inhibited the expression of PERI protein.Conclusions:Firstly, electric-acupuncture could regulate the activities and promote resynchronization of circadian rhythm effectively. Secondly, the effect of acupuncture promoting circadian rhythm resynchronization was circadian-dependent, the effect was significant at ZT12. Finally, the mechanism of acupuncture regulating circadian rhythm disturbance maybe up-regulate the expression of CKIs and phosphorylation of PER1to inhibit negative feedback.
