ObjectiveThe explore the differences of the inhibitory effect among microwave ablationcombined with endostar and ablation and endostar to provide experimental basis for theclinical use.Methodproduced New Zealand white rabbits VX2muscle transplantation tumor model byinjecting VX2organizations suspension to the left thigh muscle.②randomly divided60rabbit muscle xenograft model intofour groups: A,B,C,D. Group A is microwave ablationgroup (15),the rabbit VX-2muscle transplanted tumors was given microwave ablationtherapy,ablation conditions are40w,2min; group B,injected endostar (15),to give endostar0.75mg/Kg ear vein injection14days,inhibition of the rabbit VX-2muscle tumor growth;group C,Microwave ablation combine with endostar(20),the ablation conditions is the sameas group A and the amount of endostar is the same as group B; group D (10) for the controlgroup without any treatment. Observed tumor volume change to calculate the inhibitionrate and take the tumor for pathological examination,while using the SP method ofimmunohistochemistry,using CD34labeled microvascular of rabbit VX-2muscle-growingtumor,then give immunohistochemical staining,measured in tumor microvessel density(micro vascular density,MVD) and vascular endothelial cell growth factor (of Vascularendothelial cell growth,VEGF).Result1.tumor metastasisGroup A:two of fifteen rabbits died in three months,the survival rate was 86.7%(13/15),an autopsy found that there are metastasis,metastasis rate was53.3%(8/15).Group B:eight of fifteen rabbits died in three months,the survival rate was46.7%(7/15), theautopsy found that there are metastases,metastasis rate was60.0%(9/15). Group C: two oftwenty rabbits died in three months,the survival rate of90.0%(18/20),an autopsy found thatthere are metastasis,the metastasis rate was15.0%(3/20). Group D: ten rabbits all died inthree months,the survival rate was0%(0/10),an autopsy found that there aremetastasis,metastasis rate was100%(10/10). For the survival rate, The differences of groupD and the other three groups were statistically significant (P <0.05). The difference ofgroup B and group A,C was statistically significant (P <0.05).The difference of group Aand group C was no statistically significant (P>0.05).For the metastasis rate, Thedifferences of group D and the other three groups were statistically significant (P <0.05).The difference of group C and group A,B was statistically significant (P <0.05).Thedifference of group A and group B was no statistically significant (P>0.05).2.MVD determinationThe MVD of A,B,C and D group were19.34±4.32,22.74±3.98,10.53±2.66,37.42±3.12. The MVD of D was the largest,The MVD of A was larger than B and C. The MVD ofgroup C is the smallest.The differences of group D and the other three groups werestatistically significant (P <0.05). The difference of group C and group A,B wasstatistically significant (P <0.05).The difference of group A and group B was nostatistically significant (P>0.05).3.VEGF determinationThe VEGF positive expression percentage of A,B,C and Group D was21.52±4.47%,25.58±4.01%,12.36±3.26%,43.56±3.48%.The VEGF of D was the largest,TheVEGF of A was larger than B and C. The VEGF of group C is the smallest.The differencesof Group D and the other three groups were statistically significant (P <0.05). Thedifference of Group C and group A,B was statistically significant (P <0.05).The differenceof group A and group B was no statistically significant (P>0.05).Conclusion1.Microwave ablation caused the tumor tissue degeneration,necrosis,and inhibitionof tumor growth. But there is some reason which may render inadequatetreatment,resulting in residual tumor cells.Microwave ablation can effectively destroy thetumor microvessel,effectively inhibit angiogenesis and reduce tumor blood supply,so thatthe rabbit tumor model of tumor microvessel density and vascular endothelial growth factor is reduced.2.Endostar can reduce tumor vascular endothelial growth factor expression, inhibitionof tumor endothelial cell proliferation,migration,reduce tumor angiogenesis and reducetumor metastasis.3.Endostar combined with microwave ablation has a synergistic anti-tumor effect thansingle Endostar and single microwave ablation,it is more effective to reduce tumor vascularendothelial growth factor expression,and destruct the tumor microvascular,inhibit theformation of new blood vessels,inhibit the growth and metastasis of tumor.
