Objective:To study the changes of protein expression in rats’triceps muscle after C7nerve root rhizmotomy by two way fluorescence difference gel electrophoresis (2D-DIGE), and to explore the molecular mechanism of fuctional comensation in triceps musle.Methods:We established one-side C7nerve root rhizotomy SD rat model. Equal amounts of triceps muscle were harvested from both sides after the rhizotomy in1、2、3、6、12weeks, the normal side was designed as control group. The total proteins from the experimental side and normal side were distracted by the two way fluorescence differential in-gel electrophoresis (2D-DIGE) technique. The protein spots with statistically significant difference were screened by one-way ANOVA and identified by MALDI-TOF-MS or MS/MS.Results:1、Intensity changes of38spots were screened with statistically significant difference. And28of them were identified by MALDI-TOF-MS or MS/MS, including energy metabolism related protein, repair protein, contractile proteins, cytoskeletal proteins, elongation factor Tu of mitochondrial, calpastatin and transporters.2、The changes of protein expression in one class are similar. In one week after the rat C7nerve root rhizotomy, anaerobic metabolism enzyme, repair protein and transporters downregulated, but contractile proteins, cytoskeletal proteins and calpastatin displayed an up-regulation. In the3rd week, all energy metabolism related proteins up-regulated. Then anaerobic metabolism enzyme up-regulated, but aerobic oxidation enzymes down-regulated together with mitochondrial elongation factor Tu and calpastatin from6th to12th week. And transporters were continuing downward.Conclusions:1、Protein expressions in triceps muscle are significantly different after rat C7nerve root rhizotomy.2、Energy metabolism of triceps muscle declined in one week after the rat C7nerve root rhizotomy, and recovered in the3rd week. The functional compensation of triceps after the rat C7nerve root rhizotomy may be associated with the changes in muscle fiber types, and the regulation of this change may occur in the level of protein translation of energy metabolism enzyme. The calpain proteolytic system may take a part in it。3、The functional compensation of triceps after the rat C7nerve root rhizotomy may be associated with muscle fiber hyperplasia and suppression of degradation.
