Purpose:By using the technology of RNAi, SOCS-3gene silence in asthmatic micecan improve the inflammatory response and the balance of Th1/Th2cells in asthmas mice,which has offered the theoretical and experimental basis for the clinic.Methods:SOCS-3siRNA lentiviral vector were transfected to asthma mice by nasalinhalation.24hours after the last stimulate, get rid of one eye and collect the blood, the wayof cervical dislocation is to kill the mice and collect all of samples, then using the HEstaining、EOS staining、Immunohistochemical staining、Western Blot、RT-PCR、ELISA andso on is to observe the influence of the asthma mice and the balance of Th1/Th2cells by theSOCS-3siRNA lentiviral vector.Result:HE staining shows that the transfection of SOCS-3siRNA lentiviral vector hasreduced the inflammation and the amount of muces in asthma mice. The result ofImmunohistochemical staining、Western Blot and RT-PCR show that the express of theSOCS-3and Eotaxin are reduced compared with the normal mice. EOS staining shows thatthe EOS in asthma mice is also reduced, but the mice which are transfeced withnon-seecificity lentiviral vector are still unhealthy and have no change.Conclusion:The mice which have transfeced with SOCS-3siRNA lentiviral vector aremuch better than asthma mice, and improve the balance of Th1/Th2cells.
