ObjectiveWe detected the expression of Mismatch Reparir Protein2(hMSH2)and Bcl-2Assaciated X (BAX) protein in liquid-based cervical-cell of Negative for intraepithelial ofmalignancy(NILM),low-grade squamous intraepithelial lesion(LSIL),high-grade squamousintraepithelial lesion(HSIL) and squamous cell carcinoma(SCC),and have explored the roleand the relationship of hMSH2and BAX protein in cervical cancer and cervical cancer.Toprovide possible theory in cervical cytology to study the mechanism and the diagnosis ofcervical cancer by immunohistochemistry.Methods120cases of cervical exfoliated cytology specimens all collected from outpatient andinpatient in the Affiliated Hospital of Tai’shan Medical College during the year of2009and2011,included30cases of NILM,30cases of LSIL,30cases of HSIL and30patientsof SCC.All the specimens have complete clinical data,have no chemotherapy,radiotherapyor other target treatment before operation.Prepared three samples whether fresh or saved,which two were for hMSH2and BAXdetection by immunocytochemical,and the other was for cervical cytology diagnosis.Thepositive signal of hMSH2was localized in the nucleus, and the positive signal of BAX waslocalized in the cell cytosol.All data analyzed by SPSS12.0.The results was treated byχ2test, and we treated the correlation between hMSH2and BAX by Spearman rankcorrelation detection, and the difference was statistically significant as P<005.ResultsThe expression of hMSH2in normal cervix,LSIL,HSIL and SCC cells wererespectively16.67%,40%,60%and76.67%in liquid-based smears,and BAX were73.33%,53.33%,36.67%and20%. HMSH2and BAX protein respective expression innormal cervix and LSIL,HSIL,SCC group were also significant different (P<0.05).while the degree of cervical intraepithelial lesions increased,hMSH2protein expression wasincreased and BAX protein expression was decreased.Expression of hMSH2and BAXprotein was a significant difference (P<0.05) in SCC group,which are negativecorrelation.Conclusion(1) hMSH2protein expression increased while cervical intraepithelial lesionsincreased,and the expression of hMSH2and cervical lesions were positively correlated;BAX protein express the highest level in normal cervix, while decreased with cervicalintraepithelial lesions increased,shows there was an inverse correlation between them.(2)in SCC group,there was an inverse correlation between the expression of hMSH2andBAX protein.(3) hMSH2and BAX protein involved in cervical cancer’s occur andprogress,detection of hMSH2and BAX antibodies activity in liquid-based cervicalcytology is an effective auxiliary tool for the screening of cervical cancer and precancerouslesions,cervical lesions on clinical diagnosis.
