Objective:Screening the correlated genes of mouse lung cancer induced by tobacco smoke, using gene expression chip of mouse genome. Identifying the screening overexpressed genes by fluorescence-based Real-Time PCR, and detecting the protein expression of Ang-3in mouse lung cancer by western blot essay. To provide evidences for searching initially new correlated genes of lung cancer.Methods:1. Screening the genes of anomalous expression in mouse lung cance induced by tobacco smoke through gene expression chip of mouse genome;2. Identifying the partial results of gene chip by fluorescence-based Real-Time PCR;3. Detecting the protein expression changes of Ang-3in mouse lung cancer by western blot essay.Results and Conclusions:1. We have obtained621genes (contain293unknown function genes), including mainly oncogenes, signal transduction and path, accommodation of cell cycle, cell adhesion, immune and inflammatory reaction, cell differentiation, etc, using the gene chip of mouse genome.2. Bases on the screening results of gene chip and analyzing relative data, we piched out5genes of non-discovered currently oncogene or anti-oncogene function for analyzing their expression of mRNA in different mouse lung tissues by fluorescence-based Real-Time PCR. The expression of Ang-3、 Arg-2、H2-Ea and H2-D1in lung cancer tissues was more than in normal lung tissues(p<0.05), but the expression of Ifi203in lung cancer tissue is same as in normal(p>0.05). At last, we choosed chiefly Ang-3gene as a model to identify by western blot essay.3. The results of western blot showed that Ang-3protein expression in lung cancer tissues is significantly more than that in normal lung tissues (p<0.05), which is same with the results of gene chip and fluorescence-based Real-Time PCR.4. Based on the above evidences, Ang-3gene plays an important role in mouse lung cancer induced by tobacco smoke.
