The animal testing for the method, the rats were fed different doses of hydrolysed casein bound iron (Fe2+-Cas hydr) in fixed time and dose in everyday. Ferrous glycinate(Fe2+-Gly), Fe-deficient diet and conventional diet group were control groups. Testing Hb, RBC, HCT, FS, TIBC, TS, SOD, GSH-PX, T-AOC, MDA and the total number of bacterial colonies. To study the effect of Fe2+-Cas hydr on iron supplementation, antioxidant activity and antibacterial activity to rats. To compared the effect of Fe2+-Cas hydr in different doses of iron to rat on iron supplementation, antioxidant activity and antibacterial activity. The effect of Fe2+-Cas hydr and Fe2+-Gly in same doses of iron to rat on iron supplementation, antioxidant activity and antibacterial activity.1. To established the model of iron deficiency anemi (IDA) rat. Selected65health Sprague Dawley rats. The animals were divided into two groups, one group often which were fed conventional diet (200mg Fe/kg diet) and another group of fifty which were fed iron deficient diet (12mg Fe/kg diet). Two groups of rats had free access to diet and distilled water in plastic tank and were housed in stainless steel cage. The result show that in42days, the Hb of all the thirty rats were lower than90g/L. In45days, the Hb of all the rats were lower than90g/L. The anemia model was established successfully.2.The effect of Fe2+-Cas hydr on iron supplementation.The anemia rats were devided into five groups of ten. Three groups were fed Fe2+-Cas hydr in high, medium and low iron doses respectively. One group was control group which was fed Fe2+-Gly medium doses as well as Fe2+-Cas hydr in medium iron doses. Another group was model group which was fed nothing but iron deficient diet. The result show that After the36-day experiment, the hematology index of rats in experimental groups with high and medium doses of Fe2+-Cas hydr recovered to normal range, and no significant differences appeared in the treatment effect of Fe2+-Cas hydr and control group. Fe2+-Cas hydr with iron ranging from1.818μg/(g.d) to0.363μg/(g.d) is most effective in treating anemia.3. The research of Fe2+-Cas hydr regulated the oxidation resistance of organism. Rats serum from five groups(Fe2+-Cas hydr in high, medium, low dose. Control and model group) was used for determination of SOD, GSH-PX, T-AOC and MDA. The result show that Fe2+-Cas hydr can promote the oxidation resistance of body. As the dose of Fe2+-Cas hydr increased, the antioxidant activity were the better. When the body is in a state of anemia long time, low doses of Fe2+-Cas hydr can not reduce the degree of oxidation of the body effectively.4. The research of Fe2+-Cas hydr regulated the antibacterial of organism. All the rats dissercted, took the large intestine, take the contents from the large intestine. All the samples of the feces had been vaccinated to three kinds of selective mediums(EMB ager, MAC ager, KF streptococcus ager). The seeded plates were incubated at35℃for18-24h, then colonies count. The result show that the antibacterial activity of the Fe2+-Cas hydr in high, medium and low dose were better than model group. The higher dose of Fe2+-Cas hydr, the greater the effect is obvious.When the body is in a state of anemia long time, low doses of Fe2+-Cas hydr can not restore the body’s immune function effectively.
