| Nymphaea hybrid was imported into China as a ornamental plant at1970s, which has nine different colors including purple, blue, yellow, red and so on, so it is also called nine color lily. Some research showed that Nymphaea hybrid had the ability to reduce lipidsand anti-tyrosinase, however, there weren’t more reports about its biological activity and applicable valve, and no comparative analysis among different color. This article aimed to compare the composition, anti-oxidation,5alpha-reductase (5α-R) inhibition activities of yellow, blue and purple Nymphaea hybrid, and due to the connection between5a-R and benign prostatic hyperplasia (BPH), the article did a research about its effect on BPH. The main results are as follows:The chemical composition of Nymphaea hybrid was detected by standard method. The result showed that the Nymphaea hybrid were rich in protein, sugar, crude fiber, ash, and has17amino acids. As for water, crude fat, protein, sugar crude fiber and ash, there was few different among different colors, but as for amino acid and mineral substance, purple and blue Nymphaea hybrid was much better and valuable than yellow one.The antioxidant activity of ethanol extract were determined by DPPH assay, FRAP assay〠H2O2assayã€OH·assay, respectively. The result showed that all colors had antioxidant activity. The whole flower was better than the petal, the blue one was equivalent to the purple one, but much better than the yellow one. In addition, the correlation coefficient between anti-oxidation and polyphenol content is0.9.HPLC method were used to establish5a-R screening model in vitro:0.5mL pH6.5sodium phosphate buffer,0.1mL50%ethanol,150μL0.5mg/mL testosterone,250μL2.0mg/mL NADPH,0.5mL3.84mg/mL enzyme extract,37℃for30min, Propyl Hydroxybenzoate as internal standard. The finasteride equivalent of Yellow, purple, blue flower and its ethyl acetate, butanol and water fraction were1.48±0.15ã€1.91±0.09ã€1.99±0.08ã€3.41±0.34ã€3.83±0.11and0.40±0.24mg finasteride/g extraction. Besides, the the correlation coefficient between5α-R inhibiting activity and polyphenol content is0.9977.In BPH rats model induced by propionic acid testosterone, blue Nymphaea hybrid could reduce the rat prostate index, reduce the hyperplasia of the prostate epithelial cells, reduce the prostatic5a-R activity, reduce the PACP activity. At the same time, Nymphaea hybrid could increase the CAT activity, reduce the MDA content, and increase the GSH-Px activity.In summary, the extract from the Nymphaea hybrid had antioxidant and5a-R inhibiting activity, the purple one was equivalent to the blue one, but all better than the yellow one. In addition, blue Nymphaea hybrid could improve prostatic hyperplasia symptoms. |
