| Objective This study was conducted:(1) To detect the expression levels of IGF-1(insulin-like factor-1),PCNA,Caspase3 in normal prosatic tissues, benign prostatic hyperplasia(BPH) and prostatic cacinoma. And discuss their clinical significance. (2)To investigate the expressions of IGF-1(insulin -like factor-1), PCNA(prolifeation cell nuclear antigen) and Caspase3 in human prostatic hyperplastic tissues influnenced by 5α-reductase inhibitor on the level of protein. (3)To study the relationship between the expressions of IGF-1,PCNA and Caspase3.Mechanisims of benign prostatic hyper- plasia are studied according to aforesaid experiments.The reference for researching pathogenesis of benign prostatic hyperpastia and better treatment will be given.Methods (1) Nighty-six paraffin embedded samples of normal prostate tissues(10 cases),benign prostatic hyperplasia tissues(56 cases),prostatic cacinoma tissues(30 cases) were selected from the pathology department of the Second Affiliated Hospital of Chongqing Medical University.When the paraffin sections were made and stained by immunohistochemical PV9000 methods, detecting and analyzing the expression of PCNA,Caspase3,IGF-1 and the relationships among them in all groups.(2)Observing the expression of PCNA,Caspase3,IGF-1 and relationships among them by immunohistochemical PV9000 methods in BPH treated with 5α-reductase inhibitor, and analyzing clinic significance.Results (1) The expression of positive rate of IGF-1 in normal prostate, benign prostatic hyperplastic and prostatic cacinoma tissues respectively was: 0.073±0.012,0.116±0.025 and 0.134±0.036.Positive rate of IGF-1 in BPH group and Pca group was higher than that of normal prostatic group (P<0.05). The expression of positive rate of PCNA in normal prostate and BPH and Pca tissues respectively was: 0.113±0.139, 0.962±0.571 and 1.196±0.406.Positive rate of PCNA in BPH group and Pca group was higher than that of normal prostatic group(P<0.05). The expression of positive rate of Caspase3 in above three groups respectively was: 3.027±0.333, 2.035±0.175 and 1.304±0.304.Positive rate of Caspase3 in BPH group and Pca group was lower than that of normal prostatic group (P<0.05).( 2) The expres- sion of positive rate of IGF-1 in BPH with treated 5α-reductase inhibitor group (30 cases) and untreated group (26 cases) was:that of untreated group was 0.118±0.025,while the other group was 0.079±0.026. Positive rate of IGF-1 in treated group was lower than that of untreated group (P<0.05).The expression of positive rate of PCNA in treated group and that of untreated group was different (P<0.05). There was significant difference in the positive rate of Caspase3 in two groups. The tendency of IGF-1 and PCNA in periurethral higher than that of under amicula in treated BPH group was discovered, Caspase3 lower than that of under amicula ,but there was no statistically significant.(3)There is close correlation in IGF-1, PCNA and Caspase3 in treated BPH group(P<0.05).There is positive correlation between IGF-1 and PCNA(P<0.01), on the contrary, the expression of IGF-1 was negatively correlated with the expression of Caspase3 in treated group(P<0.05).Conclusions (1) The combining detection of the p63,IGF-1, PCNA and Caspase3 can improve the accuracy of diganosis and differential diganosis in prostatic lesions.(2)There is close correlation between IGF-1 and 5α-reductase inhibi- tor, IGF-1 probably inhibites cell proliferation and promotes apoptosis by 5α-reductase inhibitor.(3)There was difference in different situs of BPH with 5α-reductase inhibitor,which probably is cause of incompletely inhibition prostatic cell proliferation and promoting apoptosis. |
