The malignant breast cancer has increasingly become a serious threat to women’s healthand even life. The leading cause of death in patients of breast cancer is metastasis. Elucidatingthe molecular mechanisms of metastasis in breast cancer cells is critical for breast cancerdiagnosis and therapy.The previous studies in this laboratory have found that NEDD9can promote breastcancer cell invasion and migration through a process termed Epithelial-MesenchymalTransition (EMT). However, it remains unclear whether other factors are involved in thisprocess.In this paper, we observed that NEDD9induced invasion and migration in MCF-10Abreast cancer cells. Meanwhile, the MMP9expression level was significantly up-regulatedin MCF-10A cells stably transfected with NEDD9. While down-regulated MMP9by siRNAin NEDD9-MCF-10A cells, the ability of invasion and migration of MCF-10A breast cancercells was reduced. Further research discovered that the p38signaling pathway was able toinduce MMP9expression in NEDD9-MCF-10A cells.In summary, our experiments confirmed that NEDD9can up-regulate MMP9expression via the activation of the p38pathway and can promote breast cancer cell invasionand migration. The experimental data reveal the molecular mechanisms of NEDD9promotesbreast cancer cell migration, and provide a new target for the diagnosis and treatment ofbreast cancer.
