BackgroundBreast cancer is one of the most common malignant tumors of women in present, its incidence was taking on a increasing trend in our country and its onset age tend to be early. Although clinical workers engage in exploring the treatment of breast cancer with more than one hundred years, especially in the past20years, the clinical diagnosis and treatment of breast cancer had greatly improved, yet nearly more than400000people die of breast cancer a year. Like other malignant tumor, easily recurrence and metastasis was the key factor of leading to death, and also was the target of treatment. Considering it was very difficulty to control recurrence and metastasis of breast cancer, therefore, it is still necessary to make further effects to explore the molecular markers related to invasion and metastasis of breast cancer and to clarify the mechanism of recurrence and metastasis. Since that we can make the best individualized treatment plan to improve the prognosis of patients with breast cancer.EGFR was c-erbB-1’s expression product, which was one of the four members of the epidermal growth factor receptor family. EGFR was a kind of catalytic receptor with tyrosine kinase activity,which was a surface sensor crossing the membrane of the cell. EGFR can trigger a series of cascade reaction with signal transduction in intracellular when was activated. EGFR can finally activate corresponding protein and promote cell cycle from Gl phase to S phase. Existing studies have shown that EGFR plays a very important role in the process of development of tumor, such as cell proliferation, angiogenesis, metastasis and apoptosis suppression. The high expression of EGFR in solid tumor was a signal which means the diseases will further progress or indicate bad prognosis;In addition, some research have found that the tumor resistant to hormone therapy and the breast cancer lines which independent on estrogen often follow with high expression of EGFR, which indicate there was correlation between EGFR and estrogen receptor.NEDD9was found by Kuma in1992when they study the expression of some major genes which exist in the brain of embryonic period mice by subtraction cloning technology. NEEDD9’s full named is neural precursor cells expression development down-regulated9. NEDD9was also known as human enhancer of filamentation1(HEF1) or Cas-L. NEDD9was one of the members of Crk-associated substrate family,which was a skeleton protein. Recently, some research shown that NEDD9play an important role in cell growth, invasion, migration, carcinogenic transformation and so on. NEDD9was regarded as a kind of transfer marker in variety of cancers, such as:NEDD9was one of the metastasis related genes in lung cancer, NEDD9was a downstream effector molecule of cell signal transduction which involved in glioma transferred.The molecular biology of tumor research show that EGFR and NEDD9may exist a certain connection in the process of signal transduction.There was no literature reports on both of them corresponding with the evolution process of breast cancer. The expression of EGFR and NEDD9in normal breast tissue, breast benign tumor and breast cancer as well as the relations between them were study by immunohistochemical method in the experiment.The research will help us further understand the mechanism of invasion and metastasis of breast cancer so that we can provide more experimental basis for clinical diagnosis and treatment of breast cancer.Objective Detecting the expression of EGFR and NEDD9in breast cancer tissue and breast benign tumor tissue and normal breast tissue; analyzing the relationship between them and the clinicopathological features of breast cancer, as well as finding out the relationship between them, discussing both of them whether to be a index which can judge the prognosis of breast cancer and guide clinical treatment.MethodsThe clinical data of120patients with breast cancer, who were confirmed by pathological after operation at the first Affiliated Hospital of Zhengzhou University between January2010and January2012, were collected; while another42cases with breast benign tumor and normal breast cancer were used as the control group. The expression of EGFR and NEDD9in these tissue samples were detected by immunohistochemical SP method. The data was analyzed by SPSS17.0statistical software and chi-square test with P<0.05as the difference have statistically significant.Results1. The expression of EGFR and NEDD9in breast cancer tissues was68.3%and60.8%; in breast benign tumor was42.9%and35.7%; in normal breast tissue was16.7%and26.2%. The expression of EGFR in these tissue were significant difference,while the expression of NEDD9in breast cancer was significant higher than those in breast benign tumor and normal breast tissues.2. The positive expression rate of EGFR and NEDD9are both related to clinical stage, histological grade, ER, PR and lymph node metastasis (P<0.05) while nor correlated with age, tumor size, pathological type and HER-2(P>0.05).3. The expression of EGFR and NEDD9in breast cancer had significantly positive correlation.ConclusionThe high expression of EGFR and NEDD9in breast cancer correlated with axillary lymph node metastasis, TNM stage, histologic grade, ER and PR status,The expression of NEDD9and EGFR in breast cancer exist positive correlation, both of them may play an interaction role in the invasion and metastasis of breast cancer and can be a index to judge the prognosis of breast cancer and guide clinical treatment.
