ObjectiveTo observe the temporal and spatial expressions of fibroblast growthfactor receptor7、10(FGF7、FGF10)and fibroblast growth factor receptor2Ⅲb (FGFR2Ⅲb)during kidney development of mouse, and investigate therelationship between FGF7、FGF10、FGFR2Ⅲ band kidney development.MethodsThe expressions of FGF7、FGF10and FGFR2Ⅲb in embryonic(includingfetal age of12,14,16and18d) and neonatal(including the age of1,3,7,14,21and42d)mouse kidneys were examined by immunohistochemistry andImmunofluorescence. The protein content of FGF7、FGF10and FGFR2Ⅲbwere measured by computerized densitometry、stereological method andWestern blotting.ResultsImmunohistochemical and Immunofluorescence examination showed thatFGF7、FGF10and FGFR2IIIb were weakly expressed in the ureteric buds atE12d and then with the renal development, FGF7、FGF10and FGFR2IIIbwere expressed mainly in distal tubules and collecting ducts. FGF10wasexpressed also in the proximal convoluted tubule. FGF7and FGFR2Ⅲ bwasnot expressed in the proximal tubule. The renal corpuscles in eachdevelopment stage were not found the expression of FGF7、FGF10andFGFR2IIIb. The results of computerized densitometry、stereological methodand Western blot showed that from E12to N7, the expressions of FGF7、FGF10and FGFR2IIIb were increased gradually. From N7to N42, theexpressions of FGF7、FGF10and FGFR2IIIb were gradually decreased. ConclusionsFGF7、FGF10and FGFR2IIIb probably might play important roles in therenal development, FGF10is mainly in proximal convoluted tubule, distaltubules and collecting ducts and FGF7、FGFR2IIIb are mainly in distal tubulesand collecting ducts.
