| ObjectiveDental metal and its alloy have good biocompatibility. However, someresearch findings showed that metal corrosion can cause cytotoxicity, eveninjury to periodontium. Through the analysis of four types of metal extraction ofhuman gingival fibroblasts uPA and PAI-â… that the activation expressiondifferences, explore a new method to evaluate the biocompatibility of thematerial in oral repair.Methods(1) Choose healthy gingival tissue of Healthy Volunteers with writteninformed consent and primary culture of HGFs was performed using theimprovement of tissue culture method. Passage4cell were used inidentification. Vimentin and cytokeratin were examined to identify its origin andgrowth curve for observing the cell growth.(2)Make sure that the variation ofPAI-â… and uPA in Ni-Cr, Ni-Cr-Ti, Ti and rhotanium extraction byimmunofluorescence and ELISA analysis levels and relationship of PAI-â… anduPA in different metal ions. Statistic analysis were achieved by SPSS13.0software. Results(1) Vimentin stain was positive,cytokeratin and negative control group werestained negative. It was proved that the cell were fibroblasts derived frommesoblast.(2) Observe the results of fluorescent staining under electronmicroscope. The cells of Ni-Cr alloy group and Co-Cr alloy group stained darkerand equitable and have integrity morphology. In contrast the other three groupof results.(3) According to images analysis by SPSS, it shows that there is nosignificant difference inrhotanium, pure Ti alloys and control groups (P>0.05).There was also no significant difference between rhotanium alloys and pure Ticeramic alloys groups or between Ni-Cr ceramic alloys and Co-Cr ceramicalloys groups(P>0.05), but Ni-Cr ceramic alloys group or Co-Cr ceramic alloysgroup showed significant difference compared with the rhotanium, pure Ticeramic alloys and control groups in12h and24h(P <0.05).ConclusionsThe advancement of uPA and PAI-â… of HGFs induce by the corrosion ofmetal ion affect cell metabolism and extracellular matrix dissolution,even injuryto periodontium. Through the analysis of the activation expression differences ofhuman gingival fibroblasts uPA and PAI-â… in four types of metal extraction, wecan draw the conclusions as followed:rhotanium alloys and pure Ti ceramicalloys s have no obvious effect on the levels of uPA and PAI-â… ,in contrast thatNi-Cr and Co-Cr alloys showed an obvious effect on the levels of uPA and PAI-â… .Increased expression of PAI-â… and uPA was one of the factors causingdamage of HGFs and gingiva. |
