Ovotransferrin. an iron-binding glycoprotein, mainly come from birds egg white and serum. Ovotransferrin has various biological functions, such as immunomodulatory, participating in iron transportation and metabolism, antibacterial, antiviral and antioxidation etc. Ovotransferrin as functional ingredien in the health food and drug had wide development and application prospect. Investigated the immunomodulatory effects of ovotransferrin by cell cultivation in vitro and animal experiments, the main research contents and results were as follows:1. Phagocytic ability of peritoneal macrophage, MTT and ELISA methods were used to investigate effects of ovotransferrin in immunocyte. The results indicated that OVT could enhance the phagocytic ability of peritoneal macrophage(P<0.01), cooperate with LPS to accelerate the proliferation of Peritoneal Macrophages(P<0.05), significantly improve the effect of inhibition in Peritoneal Macrophage proliferation caused by CsA(P<0.05),and significantly promote the proliferation of lymphocytes(P<0.01), cooperate with LPS and ConA to accelerate the proliferation of lymphocytes (P<0.05), significantly promote the secretion level of IL-2in the normal mice splenic lymphocytes. The conclusion was that OVT was able to enhance the immunity of healthy mice immune cells in vitro.2. To understand the immunoregulatory function of ovotransferrin,its effects on spleen index, thymus index,cellular immune response and humoral immune function in immunosuppressed mice caused by cyclophosphamide were investigated. The results indicated that OVT significantly improve spleen index,thymus index, ear dwelling and antibody level in a dose-dependent manner,and all above indexes of the high dose group were higher than the model control group(P<0.01). The tested results of all of the aforementioned indexes were positive,suggesting that OVT was able to enhance the immunoreulatory function.3. The effects of ovotransferrin on peripheral blood Th1、Th2type cytokines(IFN-γ IL-4) level and the IFN-γ/IL-4ratio in immunosuppressed mice caused by cyclophosphamide were investigated. The results indicated that the IFN-γ concentration. IFN-γy/IL-4ratio in the serum of the model control group were significantly lower than the normal control group (P<0.01),and the IFN-γ concentration, IFN-γ/IL-4ratio in the serum of the high dose group were significantly higher than the model control group (P<0.01), but there was no significant difference compared with the normal control group. The conclusion was that OVT could correct the imbalance of Th1/Th2and enhance the immune function of the immunosuppressed mice.
