Objective Objective to investigate the effect of Bone marrow mesenchymal stem cells(BM-MSC)transplantation combined with tissue engineering material hyaluronic acid(HA)on the regeneration of scarring vocal cord lamina propria.Method For cultivation and identification of rat after using BM-MSC by green fluorescent protein(GFP)labeled rat BM-MSC.In vitro experiments,we detected the proliferation and expression of GFP labeled BM-MSC cells by flow cytometry,and evaluated the compatibility and stability of BM-MSC Hyaluronic embedded in hyaluronic acid(HA).In vivo experiments,16 rats were randomly divided into 4 groups.According to the right side vocal cord injury,after one month,the injection was treated with different injection,named saline group(group S),BM-MSC(group M),HA(group E),BM-MSC implantation HA(M+E group).After 1 months of right vocal cord injury,all rats were injected with normal saline(group S),BM-MSC(group M),HA(group E),BM-MSC implanted HA(M+E group),respectively,and the vocal cords were treated by injection.After 1 months of treatment,detection of type procollagen-III,fibronectin(FN),hyaluronic acid synthase-III(HAS3)and hyaluronidase(HYAL-3),smooth muscle actin(SMA)and transforming growth factor ? 1(Transforming growth factor-TGF-? 1,? 1)the expression level of m RNA,immunohistochemical evaluation of therapeutic effect;GFP marker survival BM-MSC by immunofluorescence to assess;the rats of laryngeal specimens using Hematoxylin and Eosin(H&E)after staining analysis of rat vocal fold lamina thickness at 10 magnification,the lamina propria of normal rats as control,the thickness of the inherent value of thickness,the thickest part of the thickness of the thinner part and the middle part of the thickness of the 3 values Middle value.Results BM-MSC implanted HA in vitro expressed Sca-1,hyaluronic acid receptor CD44 positive,and could continue to proliferate.In the study in vivo,the GFP labeled BM-MSC injection was still able to detect the survival of the BM-MSC after January.The levels of III-type procollagen,fibronectin and TGF-? 1 were increased in group BM-MSC of HA.Compared with BM-MSC injected alone,the expression of SMA in HA BM-MSC was down regulated,while HYAL3 expression was up-regulated.Compared with saline group,HAS3 expression was not changed,SMA expression was down regulated,HYAL3 expression was up-regulated.The thickness of the lamina propria in group BM-MSC,BM-MSC combined with HA treatment group and HA treatment group was significantly smaller than that in the control group,which was close to the normal group.Conclusions Injecting BM-MSC and HA into the damaged vocal cords of rats can promote the deposition of extracellular matrix(ECM)and growth factors,but not affect the differentiation of myogenic fibroblasts,and promote the repair of scar's vocal cords.
