Andrographis Paniculata (Burm.f) Nees has a long history of using as medicine in China, with antipyretic and anti-inflammatory, analgesic, anti-bacteria, anti-virus effectiveness. The main active ingredient in the diterpene lactones.Extraction Andrographolide active ingredients used supramolecular technology, can reach more than 50 percent of the content, This thesis is study the mechanism of enriching kalmegh active ingredients used supramolecular technology, to provide theoretical basis of Supramolecular technology enriching kalmegh active ingredients,and to provide method for Chinese traditional medicine.The main task as follows:1. Used CAChe 6.1 software system's Standard Procedure and semi-empirical MM3 field calculation method, to calculate energy and simulate the three-dimensional molecular structure of diterpene lactones/β-CD.The part of five dim lactones and the part of decahydronaphthalene link could incluse byβ-CD.2. Supramolecular body and SEA were made by Grinding. Used the HPLC to determine the content of SEA,A is 40.56% DDA is 10.89%,DA is 5.83%。A,IA,NA,DA and DDA were separated from SEA by SiO2 by column chromatography, thin layer of separation and recrystallization method,and was identified by IR,MS,1H-NMR and 13C-NMR3. The supramolecular body was prepared by saturated solution method assisted with ultrasonics. and was identified by DTA, IR, 1H-NMR. The inclusion sites was alike with computer simulation infer from IR and 1H-NMR. one is inclused the dim lactones,the other is inclused the part of decahydronaphthalene link. The IA,NA,DA'supramolecular body were firstly study in china.4. Used Uv, to determete the stability constants of diterpene lactone/β-CD inclusion compound, the results as follow:A:1052.63 L/mol, DDA:1612.90 L/mol, DA:1315.79 L/mol,NA:487.80 L/mol,IA:1612.90 L/mol. and to determete the inclusion ratio is 1:1. The inclusion compound was mixture of two kinds of inclusion compound.5. Firstly,through the study of SEA, the mixture of different content. We get the result: Ka(mixture)≈Ka(A)×A% + Ka(DA)×DA% + Ka(DDA)×DDA% +……6. Used Uv to determete the thermodynamics constant, the results as follow:△G)|─﹤0 of A,IA,NA,DA,DDA and SEA can testify: the reaction could run spontaneous and the inclusion compound is very stably, This is in line with the computer simulation results, Proved the accuracy of computer simulation...
