| Objective:To inspect the effects of adenomyosis (AM) model mice with "Pulinghuayuzhitong Formula", we observed mice uterine tissue pathology changes, matrix metalloproteinase-2(MMP-2) and tissue inhibitor of metalloproteinase-2(TIMP-2) in eutopic and ectopic adenomyosis,explored the mechanism and curative effect of "Pulinghuayuzhitong Formula"in AM, then we hoped provide more theropy basics and experimental valids for clinical treatment and further researchs.Methods:Healthy female unpregnanted ICR mice which were7weeks of age randomly divided into seven groups (blank group, model group, the "Pulinghuayuzhitong Formula"high, medium, low dose group, the control group of traditional Chinese medicine and the control group of western medicine).except the blank group,the other six groups mice were created a model of adenomyosis by Allogeneic pituitary uterine transplantation.After the model was created, continuely feed12weeks.Then mice were killed,we got the uterine specimen to inspect histological changes in each group,HE staining,SP immunohistochemical staining, eutopic and ectopic endometrial MMP-2and TIMP-2immunohistochemical determination.finally we made records, and statistical processing.Results:Compared with the blank group,MMP-2was increased and TIMP-2was decreased in eutopic endometrial,the ratio of MMP-2/TIMP-2was increased,which have significant statistical difference(p<0.01).Compared with the model group, MMP-2were decreased in eutopic and ectopic endometrial in each dose group of "Pulinghuayuzhitong Formula", the control group of traditional chinese medicine and the control group of western medicine,the high dose group was obviously decreased which has significant statistical difference (p<0.01); compared with low-medium dose groups and control group of chinese herbal,the expession of MMP-2in eutopic endometrial of high dose group of "Pulinghuayuzhitong Formula" was decreased,which has significant statistical difference(p<0.05);comared with western medicine group,the expession of MMP-2in ectopic and eutopic endometrial of high dose group has not significant statistical (p>0.05).Compared with the model group,the TIMP-2in eutopic and ectopic endometrial all increased in each dose group of "Pulinghuayuzhitong Formula", the control group of traditional Chinese medicine and the control group of western medicine,but has significant statistical difference(p>0.05).compared with each groups,the TIMP-2in ectopic endometrial has not significant statistical difference(p>0.05).Compared with the model group, the ratio of MMP-2/TIMP-2all was decceased in eutopic endometrialof each groups of "Pulinghuayuzhitong Formula", the control group of traditional Chinese medicine and the control group of western medicine.especially the high dose group descend significantly which have statistical difference (p<0.05).but the ratio of MMP-2/TIMP-2in ectopic endometrial has not significant statistical difference(p>0.05).compared with the low dose group of "Pulinghuayuzhitong Formula".the ratio of MMP-2/TIMP-2in high dose group was high decreased,which has statiscal difference (p<0.05),but has not significant statistical to middle dose group and the control group of chinese herbal(p>0.05).Conclusion:MMP-2was increased in ectopic endometrial of AM,whereas TIMP-2was decreased,the ratio of MMP-2/TIMP-2was decreased,it can improve the invade of endometrial and help development of AM.The high dose group of "Pulinghuayuzhitong Formula" can decreased the expression of MMP-2in eutopic endometrial which better than low-medium groups.compared with chinese herbal control group and western medicine group,the high dose group has not significant statistical difference.the high dose group of "Pulinghuayuzhitong Formula" can also decreased the expression of MMP-2in ectopic endometrial.but has not statistical difference.The high dose group of "Pulinghuayuzhitong Formula" can increased the expression of TIMP-2in eutopic endometrial,compared with low-medium groups,which has not significant statistical.to TIMP-2in eutopic endometrial.the high dose group of "Pulinghuayuzhitong Formula" has not significant statistical.The high dose group of "Pulinghuayuzhitong Formula" can decreased the expression of MMP-2in eutopic endometria.the action is more better than low dose group.otherwise,it can also decreased the expression of MMP-2in ectopic endometrial.but compared with low-medium groups has not significant statistical.After all.the "Pulinghuayuzhitong Formula" can inhibit the expression of MMP-2and decreased the ratio of MMP-2/TIMP-2,so it can decreased the invade of endometrial and cure the adenomyosis. |
